Object: To study the expressions of S100A4, matrix metalloproteinases (MT1-MMP, MMP-2 and MMP-9) and tissue inhibitor of metalloproteinases (TIMP-1, TIMP-2) at the gene and protein levels in cervical squamous cell carcinoma and explore the role of matrix metalloproteinases mediated by S100A4 in lymphatic metastasis of cervical squamous cell carcinoma. Methods: Reverse transcription-polymerase chain reaction(RT-PCR)and immunohistochemistry were used to detect the expressions of S100A4, MMP-9, MT1-MMP, MMP-2 and TIMP-1, TIMP-2 in 86 samples of stage Ib-IIa cervical squamous cell carcinoma treated with radical hysterectomy and systematic retroperitoneal lymphadenectomy without preoperative adjunctive therapy and 21 samples of normal cervix treated with panhysterectomy because of benign lesions at the Department of Gynecology, Shanxi Medical University First Hospital and Shanxi Cancer Hospital, between September 2003 and March 2004. Results: S100A4, MT1-MMP, MMP-2 and MMP-9 were observed in cervical squamous cell carcinoma, but all of them were extremely weakly expressed in normal cervix and the extremely higher expression level of them were detected in tumors with lymphatic metastasis than that in tumors without lymphatic metastasis. But TIMP-1 and TIMP-2 were observed in normal cervix, and both of them were weakly expressed in cervical squamous cell carcinoma, and the extremely lower expression level of them were detected in tumors with lymphatic metastasis than that in tumors without lymphatic metastasis. Conclusion: â‘ There were many kinds of MMPs in cervical squamous cell carcinoma, and this played a significant role in tumor lymphatic metastasis; â‘¡S100A4 promoted the expressions of MMPs in cervical squamous cell carcinoma, and this profited tumor lymphatic metastasis; â‘¢The advantages of tissue microarray technology: large amount of information; This could make good use of tissue resources, and could be used for immunohistochemistry and in situ hybridization and so on.
