Effects Of Troglitazone On Human Omental Preadipocytes Proliferation And Differentiation

Objective To culture human omental preadipocytes primarily and investigate the proliferation and differentiation processes in vitro .To study the effects of troglitazone on human omental preadipocytes proliferation and differentiation . Methods (1)Omental preadipocytes were isolated by collagenase type 1 digestion. We stained it with oil red O and observed the morphology for the purpose of preadipocyte identification.We observed the proliferation and differentiation process of primary cultured human omental preadipocyte.(2) This study examined the effect of troglitazone on preadipocyte proliferation.(3)This study examined the effect of troglitazone on preadipocyte differentiation by oil red O staining.(4)Values were given as means ±SE and analysed by SAS8.2 software.Differences were considered significant at a value of P<0.05. Results (1) We cultured the human preadipocyte successfully. Preadipocyte looked like fibroblast.We could see lipid droplets when preadipoctes were differentiated on the 4th day.Mostly preadipoctes were differentiated into mature adipocyte on the 21th day.(2) The effect of troglitazone on preadipocyte proliferation:1) Each group stimulated preadipocye for 48 hours,OD value indicated both 1μM and 10μM troglitazone can stimulate the preadipocye proliferation,but this effect did not rely on concentration increasing.2) Troglitazone of 10μM could enhance the preadipocyte proliferation on 48 and 72 hours.This effect was most obvious when it was stimulated for 48 hours.(3) The effect of troglitazone on preadipocyte differentiation:We could see lipid droplets become larger when cultured with 10μM troglitazone which indicated most preadipocytes were turned into mature adipocyte.Oil red O became more obvious when concentration was increased. OD value indicated both 1μM and 10μM troglitazone can stimulate the preadipocye differentiation,and the extend had the elevated trend with concentration increasing. Discussion (1) We isolated and cultured the omental preadipocytes and stimulated preadipocyte into mature adipocye.(2)Both 1μM and 10μM troglitazone can stimulate the preadipocye proliferation and differentiation.Troglitazone perhaps improve insulin resistance by increasing the number of mature adipocyte...
