Interleukin-1 (IL-1) is a kind of polypeptide growth factor which has complicated biological effect. It can be compounded and secreted by many cells. So many biological effects make IL-1 play a central role in the immune system. There are three members in IL-1 family: IL-1 alpha, IL-1 beta, and IL-1ra (interleukin 1 receptor antagonist). IL-1 gene cluster locates human chromosome 2q13-q21. It includes three correlative genes: IL-1A, IL-1B and IL-1RN. They code IL-1 alpha, IL-1 beta, and IL-1ra partitively. Different allele loci have been approved in the gene cluster which has the polymorphisms, which can lead to the transcription and influence the production and expression of IL-1. The pathogeny of cervical cancer is complex, while the susceptibility of the organism is very important. Because of the polymorphisms, IL-1 gene cluster can influence the transcription, and express different genotypes. Moreover, the frequency of the allele is different between the patients and the normal. It suggests that the polymorphisms of IL-1 may have relationship with cervical cancer. Objective: To investigate the polymorphisms of interleukin-1 (IL-1) gene cluster ,the relationship between the genotype of interleukin-1 gene cluster,and the susceptibility to cervical cancer in Chinese population. Methods: Genomic DNA was extracted from the scraped cervical epithelial cells of 50 patients with cervical cancer and 50 controls and then was amplified by PCR. The PCR product was subjected to 6% polyacrylamide denaturation gel electrophoresis and silver nitrate staining. Results: 1. The IL-1A -889C allele was detected in 0.57 of the patients and 0.71 of the controls, while the IL-1A -889T allele was detected in 0.43 of the patients and 0.29 of the controls. The frequency of C/T genotype was 0.54(27/50)in patients and 0.46(23/50)in controls. The frequency of C/C genotype was 0.30(15/50)in patients and 0.48(24/50)in controls. The frequency of T/T genotype was 0.16(8/50)in patients and 0.06(3/50)in controls. The genotype distribution of IL-1A did not differ between the patients and the controls. While the frequency of the allele T was significantly higher than it in the controls (P<0.05). 2. The IL-1B -31C allele was detected in 0.58 of the patients and 0.74 of the controls, while the IL-1B -31T allele was detected in 0.42 of the patients and 0.26 of the controls. The frequency of C/T genotype was 0.44(22/50)in patients and 0.40(20/50)in controls. The frequency of C/C genotype was 0.36(18/50)in patients and 0.54(27/50)in controls. The frequency of T/T genotype was 0.20(10/50)in patients and0.0 6(3/50)in controls. The allele frequency and the genotype distribution of IL-1B was significantly different between the patients and the controls (P<0.05). 3. In the alleles of the IL-1RN, the IL-1RN*1 allele was detected in 0.51 of patients and in 0.66 of controls. The IL-1RN*2 allele was detected in 0.46 of patients and in 0.22 of controls, while the IL-1RN*3 allele was 0.02 of patients and 0.07 of controls, and the IL-1RN*4 allele was 0.01 of patients and 0.05 of controls. There are five (1/1, 1/2, 1/3, 1/4, 2/2) kinds of polymorphism of IL-1RN were found in this study. The frequencies of 1/1, 1/2, 1/3 ,1/4, 2/2 were 0.20(10/50), 0.56(28/50), 0.04(2/50), 0.02(1/50)and 0.18(9/50)in the patients respectively, while in controls were 0.36(18/50), 0.36(18/50), 0.14(7/50), 0.10(5/50), 0.04(2/50). The IL-1RN*1 and IL-1RN*2 alleles and the frequencies of 1/2 and 2/2genotypes were much higher in the patients than in the controls (p<0.05). Conclusion: The T allele of IL-1A -889 and IL-1B -31 and theIL-1RN*2 allele have a close relationship with cervical cancer. Thepolymorphism of -31C/T of IL-1B gene and the polymorphism of IL-1RNgenotypes 1/2 and 2/2 are associated with the susceptibility of cervicacancer in Chinese. Polymorphism of IL-1B and IL-1RN gene might beused as indicators of susceptibility of cervical cancer.
