Objective: To study the protein expression of VEGF and the cerebral protective effect of Hydrosafflow yellow A in rat with focal ischemia and reperfusion, exploring the meaning of those changes and the mechanism of the protective effect.Methods: J JO male Wistar rats ageing between 3-4 month, weighting between 250-300g were used in this study, the animals were divided randomly into twenty-two groups: normal control group; sham operation group; focal cerebral ischemia and reperfusion group with intervals of 0,1,3,6, 9, 12, 24, 72, 144, 336 hours and the treatment group with Hydrosafflow A at same time points as reperfusion groups;5 rats in each group respectively. The model of the middle cerebral artery occlusion (MCAO)in rats was performed with the intraluminal filament occlusion .The reperfusion was produced by withdrawing the filament after 2 h of occlusion. The treatment groups were administed an intravenous infusion of Hydrosafflow A in the amount of 6mg/kg as soon as the MCAO was made. At each given time point,the rats were killed by the excessive anesthesia and the 10% paraformaldhyde was perfused through the aorta. The brain was cut in the coronal planes at the intervals 2mm from the front globe and the samples were handled with conventional methods. The changes were investigated by the pathological examination with the methods of HE and immunohistochemistry staining. The NMDAR 1 test box was provided by the Boster Biotechnology CO.Results: (1)HE: There was no abnormal discovery in normal and sam groups. In reperfusion groups, the neuron degeneration and edema could been seen after reperfusion 0-1 hour. In groups of the reperfusion 3-6 hour, the arrange of neurons was deeply stained of split. Some infarction can been seen in ischemic side. In the groups of reperfusion 12-24 hour, the degeneration and necrosis of the neuron was more obvious. The extent of infarction extend and the glial cells begin to proliferate. In groups after reperfusion 72 hour ,the pathological changes manifest as the infarction and the glial cell proliferate in a large number. In treatment groups, the generation and necrosis of neurons was obvious ease, and the infarction also reduced.(2)Immunohistochemistry staining: There were little endothelial cells positive expression in normal and sam groups, some value endothelial cell can be seen in creased after cerebral ischemia 2 hours and reperfusion 3 hours , and reach the top in reperfusion 24-72 hours , then decrease gradually after one week, reach normal level or little higher after 2 weeks. There were no glial cell and in normal and sam groups. Some of them can be seen increased between cerebral ischemia 2 hours and reperfusion 3 hours, and reach the top in reperfusion 9-12 hours, then decrease...
