Objective The purpose is to find serum biomarkers for laryngeal carcinoma in patient blood serum using the Surface Enhanced Laser Desorption/lonization-time of flight (SELDI-TOF) technique and bioinformatics.Methods Serum samples from 33 laryngeal carcinoma patients and 30 age- and sex-matched vocal fold polyps patients were analyzed by SELDI-TOF on a ProteinChip reader, PBSII-C. Protein profiles were generated using WCX2 protein chips. Protein peak clustering and classification analyses were performed utilizing the Biomarker Wizard and Biomarker Pattern software packages, respectively. Results The results showed that 27 peaks had significant difference between laryngeal cancer patients and vocal fold polyps patients, twelve of which were up-regulated in the patient samples, and the others were down-regulated. 3 protein peaks 2279 Da 4626 Da and 5663 Da were automatically chosen for the system training and development of a classification tree. We divided all patients by using this classification tree ,the corresponding sensitivity was 93.94% and the corresponding specificity was 100%.Conclusion The results suggest that serum is a useful resource for the detection of specific biomarkers for laryngeal carcinoma. Proteinchip Array System was a useful tool for a high throughput screening of large-sized serum samples to discover potential biomarkers for carcinoma.
