| Objective The present study was to investigate and confirm whether Rg2 has the neuroprotective effect in anoxic cultured natal hippocampal neuronal cells. The ischemic cerebral diseases is the common diseases in clinic, The epidemical researches have shown that ischemic cerebral diseases, cancer and cardiopathy have becomed one of the three most important causes of death. 70% of the survivors have different deformity, which bring about heavy burden to family and society. The classic theory believes: The Central Nervous System can not recover once injured, but with developing of the science and technology and Molecular Biology , many scholars have affirmed that the Central Nervous System can also recover once was given proper conditions. That makes people to be aware that the prevention and therapy to ischemic cerebral diseases is workable. The present study emphasized on: The protective effects and mechanism of the Rg2 on hippocampal neuronal cells against anoxic .Methods Neurons in hoppocampal region of 1 -3d newborned Wister rats were cultured by cytosinc arabinoside (Ara-c)-supplementing method, The neurons of the 10-14d were cultured for 1h with 95%N2+5%CO2.Different dosage of ginsenoside Rg2 was given after anoxic for 4h to prevented therapy, consrasting with nimodipine. The activities of the neurons was tested by trypan blue method. The apoptosis rate and death rate of hippocampal neuronal was tested by using flow cytometry (FCM). The concentration of cellular free calcium in the cells was tested by Fluorescence spectrophotometer(Fura-2). The morphological changes was observed by Giemsa .The degree of the apoptosis was tested by DNA Ladder. The contents of MDA and NO and the activities of SOD in the supernatants of cells were determined by bicchemical methods. Results Rg2 significantly increased the activities of hippocampal... |
