Objective To study the expression of hypoxia-inducible factor-1 alpha (HIF-1 α), vascular endothelial growth factor (VEGF) and insulin-like growthfactor-I (IGF-Ⅰ) in endometrial carcinoma and investigate the relation between HIF-1 α , VEGF and IGF-Ⅰ.Methods Expression of HIF-1 α , VEGF, IGF-Ⅰ protein were assessed by immunohistochemistry method (PicTureTM method) respectively in 10 cases of atypical hyperplasia of endometrium and 42 cases of endometrial adenocacinoma and compared with clinical characteristics, such as age, clinical-stage and pathological grade. 20 case of normal endometrium tissues were investigated as control group.Results 1. HIF-1 α , VEGF and IGF-Ⅰ was expressed in endometrial adenocacinoma tissues, the positive rate in endometrial carcinoma were 64.29%, 61.90% and 59.52%. The positive rate in atypical hyperplasia were 20%, 30% and 10%. In contrast, normal endometrium did not express HIF-1 α , VEGF and IGF-Ⅰ. The expression of HIF-1 α , VEGF and IGF-Ⅰ protein in endometrial adenocacinoma was significantly higher than in normal endometrium tissues (X2=25.261, X2=22.094 和X2=24.239, p<0.01 ) .2. The positive rate of HIF-1 α was strongly associated with the clinical-stage. The expression rate of HIF-1 α protein in stage Ⅰ, Ⅱ and Ⅲ were 42.11% , 81.82% and 83.33% ( x2=7.435, P =0.006 ). There was no relationship between HIF-1 α protein expression and age or histological grade (P>0.05) .3. The positive rate of VEGF was strongly associated with the clinical-stage . The expression rate of VEGF protein in stage Ⅰ, Ⅱ and Ⅲ were 36.84%, 81.82% and 75.00% (x2=6.578, P =0.010 ) . There was no relationship between VEGF protein expression and age or histological grade (P>0.05 ).4. There was no relationship between IGF-Ⅰ protein expression and age or clinical-stage or histological grade ( P>0.05 ).5. The relation between HIF-1 α , VEGF and IGF-Ⅰ: The expression of HIF-1 α was positively associated with VEGF (r=0.469, x2=9.241, p=0.002) , The expression of IGF-Ⅰ was positively associated with VEGF ( r=0.452, x2=8.576, P=0.003 ), but there was no relationship between HIF-1 α and IGF-Ⅰ ( r=0.209, x2=1.842, P=0.209).
