COX, which is restriction enzyme of biologic synthetic reaction of PGE, has two types: one is COX-1, another is COX-2; COX-1 is structural gene ,and take part in "guarding family" . When the cells keep in quiet ,it isn't easy to check COX-2 , But if there is inflammation , stimulated by mitogen ,the COX-2 mRNA increases quickly .The stimulation factors exist in or out of cells including : growth factors and cell factors (such as PDGF PAF ,TNF, TGF, EGF, bFGF , IL21 ), TPA , oncogene ,ET, NO and so on .in according of antinflammation and analgesia , COX-2 inhibitor is not revolution of treating arthritis , its real contribution is that: in recent years it has been found that the expression of COX-2 exists in a lot kinds of tumors ,and participate in tumors' occurring, growth and metastasis .The important affection of COX-2 in carcinoma is correlated to affecting mitosis ,enhancing cell's adhesion preventing tumors growth and lowering immunologicsurveillance. Now , COX-2 is marker of tumor and new target of chemoprevent treatment in carcinoma.Metastasis tumor of bone is taking place ,when malignanttumor of body transfers to bone trough blood vessels. Today ,Tumor is the first disease to threaten human health researches show that 70% tumors can metastasis to bone . With the development social economic and medical technology ,more and more patients with MTB need to be treated ;Now sugery, chemotherapy and radiotherapy can ease pain and improve quality of life, but it still doesn't solve how to extend the life of patient with MTB . Today researches show that many kinds of tumors have high expression of COX-2 ,and that COX-2 maybe take part in tumors occurring , proliferate and metastasis , but there is still no report about COX-2 expression in MTB . We collected paraffin specimen of MTB , and sectioned every other 4-6 um . By IH and ISH ,we found that COX-2 protein and mRNA has high expression in MTB , The positive rate of COX-2 in MBT is 62.1%(36/58) . The super expression of cyclooxygenase-2 protein and mRNA was observed in MTB. The expression of cyclooxygenase-2 had no correlation to the cancer clinic X-ray types, original focuses and sites of MTB.Because COX-2 has important affection in treatment of tumor, we have produced special select COX-2 inhibitors which can treat carcinoma and have no side effect in gastrointestinal tract . such as Celecoxib , it can prevent 93% experimental animas occurring carcinomas of colon , prior to Asprine .
