ObjectiveThis study was designed to investigate the effect of different concentrations of indocyanine green (ICG) on the retinal histology and morphology in porcine eyes. Our objective was to provide theoretic base of the best concentration of ICG used in internal limiting membrane peeling operation.MethodsSixty-five porcine eyes were used within 5 hours after enucleation. The eyes were hemisected and the vitreous were removed. We can get the normal retinal structure from the five eyes as normal group. The rest can be divided into three groups: A, B, C for 20 eyes respectively. 1%, 0.5% and 0.05% ICG (0.5ml) were applied over the macula. After 60 seconds the ICG solution was drained away. Then, the complete eyecup was irrigated and filled with fresh BSS. The posterior pole was illuminated with a standard surgical light pipe and light source at maximum power for 3 minutes. The resected retinas were fixed and frozen to specimens, then observed by light microscope .ResultsIn group A (0.05% ICG): the retinas of 18 porcine eyes showed no damage on histological structure. In group B (0.5% ICG), 16 retinal structures were destructed and edema of fibers layer, ganglion cell layer and inner plexiform layer can be observed.
