Aim: Asymmetric dimethylarginine(ADMA) is an endogenous inhibitor of nitric oxide synthase(eNOS), which can compete against L- Arginine for binding the active site of NOS, and reduce the synthesize of NO. ADM A increases in the plasm of patients with hyperlipidemia, hypertension or diabetes, which can also be found in animal models. So, ADMA is considered as the risk factor of cardiovascular diseases. Xue Zhi Kang capsule is a new Chinese medicine that is manufactured according to the web theory of Chinese medicine, clinical tests demonstrated that Xue Zhi Kang capsule relieved cardiac angina and reduced blood lipid significantly. Animal experiments also demonstrated that this drug clearly reduced the blood lipid of hyperlipidemia rabbits that were induced by food, while little is known about the effects of Xue Zhi Kang on plasm ADMA. In this experiment, hyperlipidemia and atherosclerosis were induced by 4 weeks high cholesterol diet, and the effects of several drugs on plasm ADMA were tested through biochemical analysis of blood preparation and pathological sections observation.Method: 24 male new west white rabbits were used , they weighted 2±0.2 kg, and were divided into 4 groups randomly: 1) the normal control group that was fed with common feed, 2) the hyperlipidemia group that was firstly fed with 100g hyperlipid feed (The feed contained 1% cholesterol, 7% egg, 12% Lard, 80% common feed), then fed with common feed. 3) Xue Zhi Kang capsule group was fed with hyerlipid food together with l.Og/kg Xue Zhi Kang capsule, 4) Shu Jiang Zhi groupwas fed with hyerlipid food together with 1 table/rabbit Shu Jiang Zhi table. All rabbits were caged singly, water was available for all groups. The animals were weighed once per week and fed for 4 weeks. The dosage was adjusted according to the weight. Blood specimens were collected by the central arteriole of the rabbits' ear after fasting. The animals were anesthesia by pentothal sodioum (10mg/kg), aorta specimens were collected for further experiment. Biochemical analysis for the blood: 1) Enzyme assay method was used for detecting total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C) , low density lipoprotein cholesterol (LDL-C) .2) Chromatometry was used for plasm dialdehyde (MDA) detection. 3) Asymmetric dimethylarginine( ADMA) was detected by high liquid chromatography . Pathological sections of aorta were embedded in paraffin, stained with HE and observed by microscope.Result: The cholesterol (CHOL)and low density lipoprotein (LDL-C) of the Xue Zhi Kang capsule group and the Shu Jiang Zhi group was significantly lower than that in the hyperlipidemia group. The CHOL and LDL-C containment of each group was as follows: the Xue Zhi Kang capsule group: CHOL(800.8±144.6mg/dl), (651.8±131.1 mg/dl), the Shu Jiang Zhi group :CHOL(397.8±197.6mg/dl), LDL-C(651.8±166.6mg/dl), the hyperlipidemia group: CHOL(2578±561.9mg/dl), LDL-C(2063±630.1 mg/dl), the normal control group CHOL(26.37±10.82mg/dl), LDL-C(9.000±3.545 mg/dl).The asymmetric dimethylarginine of the Xue Zhi Kang capsule group and the Shu Zhiang Zhi group was also lower than that in the hyperlipidemia group. The plasma ADMA of the Xue Zhi Kang capsule group, the Shu Zhiang Zhi group, the hyperlipidemia group, the normal control group was 163.3±57.4 μ mol/L, 223.1±11.2 μ mol/L, 456.0±215.6 μ mol/L, 80.4±92.6 μ mol/L respectively The adipose cells in the pathological sections of the hyperlipidemia group can observed clearly under the endothelium of the thoracic aorta, while they can not be found in the drug intervention groups.Conclusion: Asymmetric dimethylarginine(ADMA) is an endogenous inhibitor of nitricoxide synthase(eNOS), previous envidences shown that ADMA increase in the plasm of patients with hyperlipidemia, hypertension or diabetes,, ADMA may be a new risk factor for cardiovascular diseases.
