Objective: to explore the effect of TNP470 and Gemcitabine on human pulmonary adenocarcinoma cell A549 and umbilical vein endothelium cell ECV-304 to inhibit the expression of VEGF, FLT-1, KDR.Methods: The effect of TNP470 and Gem to the proliferation of A549 and ECV-304 and their toxicity were mensurated by MTT after stimulated with them seperatedly. Semi-quantitative RT-PCR and Western—blot were used to estimate the influence of Gem and TNP470 to the expression of gene and protein of VEGF , FLT-1 and KDR after treating the cells with both Gem and TNP470 or seperatedly in different concentrations.Results The proliferation of A549 and ECV-304 were inhibited by Gemcitabine and TNP470 in vitro(GemlC50 :10-4 mg/ml, TNP470IC50:10-2 mg/ml), TNP470 combined with Gemcitabine can down-regulate the expression of mRNA and protein of VEGF, FLT-1 and KDR markedly (p<0.05).Conclusion TNP470 combined with Gemcitabine can inhibit the expression of VEGF, FLT-1 and KDR and TNP470 can cooperate with Gem anti-tumour.
