The influence that electric acupuncture pretreatment done to the formalin caused pain rat's painful behavior response, the different gene's expression and NOS.Objectives:Through found the formalin pain rat model, adopt the pre-disposal methord of electric acupuncture"zu san li"and"huan tiao" to observe each rat's painful behavior response ,measure the difference gene manifestation and apply immunohistochemistry staining methord meansure the content of NOS in rat's dorsal thalamus during the different time. From the spinal cord, thalamus level and animal ethology angles to investigate the internal mechanism of acupuncture's accommodative function, further and deeply study the mechanism of how acupuncture to cure ache.Methods:(1) Electric acupuccture rat's "huan tiao ", "zu san li "as pretreatment; (2)Adopt injecting the Formalin solution into the right postal paw of rat to made the model of pain; (3)Adopt injecting physiological saline into the right postal paw of rat to made the control group; (4)through record the times of the rats lick to right postal paw to meansure the painful behavior response; (5)Make use of the AP- PCR to observe ache difference gene expression condition; (6)Adopt the immunohistochemistry staining methord meansure the content of NOS in rat's dorsal thalamus .Results: (1)The rats which injected the physiology brine or the formalin all appear to the behavior of lick the claw in 0-10 minutes and 20- 40 minutes, the rats which injected the formalin increased the times of lick to paws with the time extended , but The rats which injected the physiology brine decreased instead. After electric acupuncture pretreated the rats which injected physiology brine or formalin all lessened the time of lick the paws compared with the none acupuncture gorup; (2) The random primer AP- PCR expanded outcome of each experiment gorup formed several band without complete repetition between each group; (3)the rat did'nt injected the physiology brine and the formalin NOS male nerve cell's endochylema stained thinner, apophysis number is less, the quantity of the cell is little; injecting the physiology brine or the formalin the male nerve's endochylema apophysis dyed thicker ,apophysis number is the most, the quantity of the cell is larger; after acupuncture per-treated,the rat injected the physiology brine and the formalin NOS male nerve cell's endochylema stained thick, apophysis number is more, the quantity of the cell is large.Conclusion:The electric acupuncture per-treatment can activate the anti-ache system of the spinal cord and up spinal cord level, raising the pain threshold of the rat, easing response oflick the claw stimulated by the physiology brine and the formalin; The electric acupuncture per-treatment can reduce the expression of the NOS in rat's dorsal thalamus ,decrease the NO,enduce central analgesial function .
