This dissertation includes six parts . Chapter 1 Documents about the research progresses in the chemical constituents andpharmacological activites of Schizonepeta tenuifolia Briq. in last twenty years were summedup and analyzed, 35 references were cited. Chapter 2 To optimize the technology of extracting chemical constituents from thespikes of Schizonepeta tenuifolia Briq. With hesperidin and total flavonoids as criterions,the water extraction and ethanol extraction technologies of extracting the constituents fromthe spikes of Schizonepeta tenuifolia Briq. were compared by HPLC and VIS spectrophoto-metry;and the L9(34)orthogonal experimental design was used to optimize the conditions ofthe ethanol extraction technology. As a result, the conditions of the ethanol extractiontechnology of extracting the constituents from the spikes of Schizonepeta tenuifolia Briq.were A2% ethanol, C1 hours of reflux, D2fold solvent and B3 times of extraction. Observing the absorption faction with the amount of the total flavonoids and hesperidinof the spikes of Schizonepeta tenuifolia Briq. on the five kinds of macro absorption resins. Wedraw the conclusion that the A resin has the best absorptive ability on the separation andenrichment of the effective fractions from the spikes of Schizonepeta tenuifolia Briq.. Thebest technology term is that the extraction substance with C2 medicinal material/ml densitypassed the resin column that d/l is B2 with the D1 absorbed speed, washed out with A2%ethanol for 5 times of resin volume with E3 eluted speed. Chapter 3 10 compounds were isolated from the effective fraction of the spikes ofSchizonepeta tenuifolia Briq.(Labiatae) by repeated silica colum chromatography, RP-18silica colum chromatography and Sephadex LH-20 colum chromatography. Based on thespectral data(1D, 2D NMR, MS),10 of them were characterized as luteolin (JS-1) ,apigenin(JS-2),anti-cinnamic acid(JS-3),β-sitosterol (JS-4),ursolic acid(JS-5),hesperidin (JS-6),3-hydroxy-4(8)-ene-p-menthane-3(9)-lactone(JS-7),tilianin(JS-8), hesperitin-7-O-glucoside(JS-9) and schizonepetosides B(JS-10). Among them, compounds JS-8 and JS-9 were firstisolated from this plant. Chapter 4 Establish methods of determinating the total flavonoids in the effectivefractions of the spikes of Schizonepeta tenuifolia Briq. with the evaluation quota of luteolin.The calibration curve is linear at ranges of 0.1752~1.0512mg (for luteolin ,r=0.9998). Theaverage recoveries were 98.78%(RSD=2.02%,n=6). The contents of index compounds including luteolin and hesperidin are determined byHPLC. The calibration curve are linear at ranges of 0.036~0.12|ìg(for luteolin ,r=0.9999),0.27~0.9μg(for hespridin, r=0.9999) respectively. The average recoveries are 100.93%(RSD=1.59%, n=6) and 98.15%(RSD=0.43%, n=6) respectively. Chapter 5 The investigations on the chemical constituents of stems of Schizonepetatenuifolia Briq.were shown in this chapter .As a result,9 compounds were isolated from theeffective fraction of the stems of Schizonepeta tenuifolia Briq.(Labiatae) by repeated silicacolum chromatography, RP-18 silica colum chromatography. Based on the spectral data(1D,2D NMR, MS), 9 of them were characterized as ursolic acid(HJ-1),2, 6-dimethoxybenzoquinone(HJ-2),anti-cinnamic acid(HJ-3),schizonepetosides B(HJ-4),β-sitosterol(HJ-5) , hesperidin (HJ-6) , 2α,3β, 24-trihydroxy-olean-12-ene-28-oic acid (HJ-25) ,daucosterol(HJ-28),2α,3α, 24- trihydroxy-olean-12-ene-28-oic acid (HJ-29). Among them,compounds HJ-25 and HJ-29 were new compounds, HJ-2 was first isolated from this plant. Chapter 6 "The finger prints" of effective fractions of spikes of Schizonepetatenuifolia Briq.and effective fractions of stems of Schizonepeta tenuifolia Briq.wereestablished by HPLC.
