Objective:The purpose of this study was to investigate the expression of HGF and E-Cadherin in farcancer lung tissue , paracancer tissue and lung cancer tissue to evaluate the relationship between these two factors and lung cancer. And to explore the role and chinical and pathological significance of HGF,E-Cadherin in lung cancer tumorigenesis and tumorprogress.AgNOR was also used to find the relationship with these two factors. Methods:The two-step immunohistochemical method was used to detect the expression of HGF,E-Cadherin in all 30 cases of farcancer lung tissue and paracancer tissue ,48 cases of lung cancer was also detected.And the relationship between the expression of HGF, E-Cadherin and histology type,differentiation degree,lymph node metastasis and clinical stages. The SPSS10.0 statistic software package for Windows was used to perforn the analysis. Results 1.The expression of HGF: ①The positive rate of HGF was 23.33%(7/30) in farcancer lung tissue and 36.67%(11/30) in paracanccer tissue and 66.67%(32/48) in lung cancer tissue. Statistic analysis showed that the expression of HGF was not significantly different between farcancer lung tissue and paracancer tissue (x~2=1.270,P>0.05).But the expression in farcancer and lung cancer was significantly different (x~2=0.010,P<0.05),paracancer and lung cancer was also significantly different (x~2=13.867,P<0.05).②The positive expression rate of HGF in different histology type was that 54.17%(13/24)in squmaous cell carcinoma(SCC),71.43%(10/14) in adenocarcinoma(Ad), and 90%(9/10)in small cell lung cancer(SCLC).Statistic analysis showed that the expression of HGF had no significant difference in SCC and Ad(x~2=1.103,P>0.05).And there was no significantly different between SCC and SCLC(x~2=2.555,P>0.05). And there was no significantly different between Ad and SCLC either(x2=0.354,P>0.05).③The positive rate of HGF in different histology differentiation was 55.17%(16/29) in high (middle) differentiation group, 84.21%(16/19) in none(low) differentiation group. Statistic analysis showed that the positive rate of HGF was significantly different between high (middle) differentiation group and none(low) differentiation group (x~2=4.356 , P<0.05).④The positive expression rate of HGF in different TNM stages was that 42.11%(8/19)in stageⅠ, 82.35%(14/17) in stage Ⅱand 83.33%(10/12) in stage Ⅲ.Statistic analysis showed that the expression of HGF had no significant difference between stage Ⅰand stage Ⅱ(x~2=6.116 , P<0.05).But there was significantly different between stage Ⅰand stage Ⅲ(x~2=5.134,P<0.05). But there was no significantly different between stage Ⅱand stage Ⅲ(x~2=0.005,P>0.05). ⑤The positive expression rate of HGF was 84.62%(22/26), 45.45%(10/22) in lung cancer tissue with or without lymph node metastasis.Statistic analysis showed that the positive rate of HGF was significantly different between lung cancer tissue with or without lymphoglandula metastasis(x~2=8.224,P<0.05). ⑥The HGF positive and negative group had a different number of AgNOR,in HGF positive group the x ±s was 7.309±1.113 and in HGF negative group it was 5.634±1.247.And there was significant difference between the two groups. 2. The expression of E-Cadherin: ①The positive rate of E-Cadherin was 73.33%(22/30)in farcancer lung tissue and 66.67%(20/30) in paracancer tissue,and 35.42%(17/48)in lung cancer tissue. Statistic analysis showed that there was no significantly different between farcancer lung tissue and paracancer tissue (x~2=0.317,P>0.05). But there was difference between paracancer tissue and lung cancer tissue(x~2=7.231,P<0.05);and also a significant difference between farcancer tissue and lung cancer tissue was found(x~2=10.617,P<0.05). ②The positive expression rate of E-Cadherin in different histology type was that 33.33% ( 8/24 ) in squmaous cell carcinoma(SCC),35.71%(5/14)in adenocarcinoma (Ad) and 40.00%(4/10)in small cell lung cancer (SCLC).Statistic analysis showed that the expression of E-Cadherin had no significant difference in SCC and Ad(x~2=0.022,P>0.05).Andthere was no significantly different between SCC and SCLC (x~2=0.137,P>0.05). And there was no significantly different between Ad and SCLC either(x~2=0.046,P>0.05). ③The positive rate of E-Cadherin in different histology differentiation groups was 48.28%(14/29) in high (middle) differentiation group and 15.79%(3/19) in none(low) differentiation group. Statistic analysis showed that the expression of E-Cadherin had a significant difference between the two groups (x~2=5.296,P<0.05). ④The positive expression rate of E-Cadherin in different TNM stages was that 57.89%(12/19)in stage Ⅰ,17.65%(2/17)in stage Ⅱand 25.00%(3/12)in stage Ⅲ.Statistic analysis showed that the expression of E-Cadherin had a significant difference between stage Ⅰand stage Ⅱ(x~2=9.972,P<0.05).And there was also a significantly difference between stage Ⅰand stage Ⅲ(x~2=4.288,P<0.05). But there was not significantly different between stage Ⅱand stage Ⅲ(x~2=0.864 , P>0.05). ⑤The positive expression rate of E-Cadherin were 19.23%(5/26) and 54.45%(12/22) in lung cancer tissue with or without lymphoglandula metastasis.Statistic analysis showed that the positive rate of E-Cadherin was significantly different between lung cancer tissue with or without lymph node metastasis(x~2=6.497,P<0.05). ⑥The E-Cadherin positive or negative group had a different number of AgNOR,in E-Cadherin positive group the x ±s was 6.095±1.315 and in HGF negative group it was 7.184±1.030.And there was a significant difference between the twogroups(t=3.175,P<0.05). Conclussions 1.The expression of HGF in farcancer tissue and paracancer tissue have no significantly different,but there were significantly different between farcancer and lung cancer group,between paracancer and lung cancer group.We can say that HGF was a promotor in the tumorigenesis in lung cancer.Since there was no evident relationship between farcancer and paracancer,we might say that the expression of HGF was not an early accident in tumorgenesis. 2.The expression of HGF was different in differentiation degree,lymphoglandula metastasis and TNM stage.But there was no significant difference between pathological type.There was no significant difference between TNM stage Ⅱand Ⅲ. 3.HGF positive or not had some relationship with the number of AgNOR,which suggested that the HGF positive cells were more active in cell growth. 4.E-Cadherin expression rate was different between lung cancer and paracancer or farcancer,which meant that the loss of E-Cadherin helped in tumorgenesis.But it might not be an early accident since there was no difference between farcancer and paracancer group. 5.There was no evident relationship of E-Cadherin among different pathological types.But a significantly different with differentiation degree was found,the high(middle) differtiation group had a higher expression rate than the none(low)...
