Abstract We use immunohistochemical technique to examine the expression of PCNA and image analysis technique to study the DNA content,AgNOR counts in 67 case patients of primary liver cancer. The results showed that PCNA immunoreactivity was confined to the nuclear of cancer cells.The labelling index ranged from 4.3-90.5%(median:33.68%).PCNA Li can devide into three grade:Ⅰgrade(0-25%)21cases,Ⅱgrade(26-75%)43cases,Ⅲgrade(76-100%)3cases.PCNA LI in lower differentiation,middle differentiation,higher differentiation is:61.6±16,38,36.5±7.87,10.7±7.12。The PCNA LI showed a correlation with size of tumor, differentiation and prognosis of patients and no relationship with age,gender. DNA content and AgNOR counts also have relationship with differentiation of tumor and prognosis of patients. Furthermore, the expression of PCNA significently correlated with DNA content and AgNOR counts in primary liver cancer. The results suggest that a combining study of these three indexes can be more accurate to detect the proliferating stages of cancer cells and of great value in assessing biological behavior and the prognosis of primary liver cancer.
