Macrophages, cells participating in immunomodulation, play an important role in host nonspecific and specific immunological functions. Categorzied into antigen-presentation cells, monocytes/macrophages have the potential of antigen presentation besides phagocytosis, assiting the activation of Tlymphocytes and stimulating each other for immunomodulation. Also, macrophages are important effector cells in antitumor immunity. In addition to the profound selectivity in killing tumor cells with most normal cells unharmed, macrophages' effects have little to do with tumor antigens and tumor cell proliferation and are significant to those tumor cells resistant to chemotherapy or radiotherapy too. Only activated macrophages can produce antitumor effects. IL-1 accelerates the production of antibody in Blymphocytes cells. Tlymphocytes express IL-2 receptors, produce massive IL-2, and improve the cytotoxicity of NK cells. TNF-a and NO are two major tumorcidal effector molecules of activated macrophages. Studies by Klostergaard et al. have revealed that there exist at least two macrophage tumorcidal mechanisms including TNF-a and NO pathways and they interact synergistically. TNF-a functions by inducing tumor cell death, inhibiting tumor angiogenesis and causing tumor hemorrhagic necrosis. NO provokes inactivation of rate-limiting enzyme and ribonucleotide reductase for DNA synthesis in aconitase of tricarboxylic acid cycle and finally results in death of tumor cells. INF-y exhibits strong effects on viral particles and tumor. It induces the expression of MHC II antigens such as monocytes, macrophages and dendritie cells and leads them to participate in antigen presentation and specific immunological recognition. INF-y cooperates in inducement of TNF and enhancesmacrophages' effects on pathogenic microorganisms. The anti-tumor mechanism of IFN is that it can inhibit the proliferation of tumor cells, induce NK and CTL to kill tumor cells, cooperate with IL-2 to enhance the activity of LAK. and induce tumor cells to express MHC II antigens so to improve the sensitivity to tumor cells.NCPP is an average grain without formaldechyde which is lower pyrogen, better absorbency.It can reduce CPP's side -effect such as fever, chestache, ejected part turgescence, callosity, liver damnification and so on.Search results show that NCPP is a worldwide innovation. We have applied for a national invention patent (No. 02123571.6) and are expected to own the intellectual property right completely.Methods:NCPP is a new type of biological response modifier that is attained after crushing CP by nano scale technology, wiping off invalid portion and collecting deposition.Splenic index was measured at the day of 14 after mice were injected peritoneal with NCPP or CPP. Ehrlich's ascites carcinoma cells were injected into the peritoneal cavity of mice and NCPP or CPP was given in the same way the day before the injection and every other day thereafter for totally 5 times. The survival rate was figured out. At the day of 10 after the injection of NCPP or CPP, a series of measurement were made to obtain items including the phagocytic indext and rate, the production of hydrogen peroxide and nitric oxide by macrophages, splenic NK cell activity, and the proliferation index of T cells.Results:Both NCPP and CPP prolonged the survival of tumor-carried mice. They all induced a high splenic index, a significant increase in the activity of macrophage (P<0.01) and splenic NK cells (P<0.05), but not in the proliferation index of T cells (P>0.05). There was no significant difference between NCPP and CPP for alltests. NCPP showed the same effects in tumor inhibition and splenic stimulation as CPP that were achieved primarily through the activation of macrophage and NK cells.Conclusions:NCPP can activate immunocytes to excrete diversity of cellular factor and signification medium which involved macrophage, NK cell, etc.the result of experiment indicate that NCPP can enhance body immunity effectively, for example accelerating macrophase multiplication, increasing macr...
