The neuromuscular junctions (NMJ) of frog and toad are often used as a model for a study of mechanisms underlying synaptic transmission. Early in 1952, Fatt and Katz first recorded spontaneous miniature end-plate potentials(MEPP) in frog NMJ, and showed that they were the result of spontaneous release of transmitter substance, acetylcholine (ACh), from motor nerve terminals. Two years later, Katz announced that hypothesis of quantum of release of transmitter was conjectured. He thought that the disperse mutation of MEPP is the result of coordinate quantity of ACh molecule, which is called quantum, the elemental unit of release of transmitter. But along with the development of research, hypothesis of vesicle met variouschallenge of experiment event. Some pursuer aptly amend it. Isreal and Taue submit the hypothesis of vesigate. They supposed that there was a construction depending on Ca2+ and combined ACh, which is called vesigate. When vesigate was open, it was releasing ACh from serum. Many new methods have been created along with kinds of appliance of new techniques after the research of half century. Instruments and materials of these methods are mostly very expensive, and processes are to be more and more complex. These ones are quite indispensable for the research of probing deeply into mechanism of spontaneous release. But it may be asked that if we must pay so much when we use existing outcome to researching influence on spontaneous releasing by new drugs. Before we mostly observed 2 targets of MEPP, amplitude and frequency. Gathering enouph specimens, we took an ulterior step which delt with these specimens statistically, and we found that there are many clues of spontaneous transmitter release in the phenomenon. Our group' s research before showed that Tripterygium Wifilfordii Polyglycosidium (TWP) could enhance the amplitude of MEPP. The detail of the mechanism is unknown. Tripterygium Hypoglaucum Hutch (THH) is in the same category of TWP. Root ofTripterygium Hypoglaucum (THR) was used to cure the auto immunity disease too. It has not been reported if THR has analogous working by TWP. Dragon Blood has curative effects that makes the blood system dynamic, spreads out the gore, makes the wound constringent, and makes the muscles strong. Stauntonia chinensis (DC) is used principally to cure all kind of neuralgia and rheumatism. These three drugs have not been elaborated the mechanism of remedy through MEPP. We will show the quest in this way in the article. And then, in the experiment we discovered the two original indexes of MEPP ( amplitude and frequency ) could been dealt in some way, and these methods have not reported before. We think they would be help of researching the mechanism of spontaneous transmitter release. For better apprehension, these methods will put in the former to introduce.The results and conclusion are as follows: 1. In the article we' ve found a way to hypothesize the location that drugs act on the NMJ, which is base on the principle that different location influence can make the index of MEPP show different integrate exhibition. When we take this way to research we must notice them: first, stimulating the sample in low frequency andintensity sostenuto, in order to ensure that Cf can be seperated from Cb; second, when we deel with the index statistically, we can' t make the rank few, cause Cf will submerges. You' d better have 20-25.2. It is supposed that TWP can enhance emission quantity of vesigates, and the action locality is on vesigates. It is supposed that THR can enhance emission quantity and opening velocity of vesigates, and the action locality is on vesigates.It is supposed that DB can reduce the number of vesicles, enhance the opening efficiency of AChR channel, And make the aggregation less apparent. The action localities may be presynaptic terminals and AChR channels. It is supposed that DC reduce emission quantity of vesigates, then make emission of vesicles more active, and the action locality is on presynaptic membrane.3. Vesica is unlikely to be released...
