To study the effects of REM (rapid eye movement ) sleep deprivation recoveryand drug intervention on learning and memory ability and the expression of the number of neuron nitric-oxide synthase positive cell and mRNA in rat cortex and hippocamp. The results showed that the learning and memory ability of sleep deprivation groups are decreased compared with CC group and TC group. The longer of sleep deprivation, the lower of the learning and memory ability and the more of the number of positive cell of nNOS .The longer of sleep recovery and the higher of the learning and memory ability and the fewer of the number of positiveuline cell of nNOS. The RT-PCR results supported these observations.The drug of XNJ can lower the effect on memory casused by sleep deprivation and relatively decrease the number of positiveuline cell of nNOS .The results suggested that REM sleep deprivation can increase the number of positive cell of nNOS, nNOS generate a lot of NO.NO has toxic effect on cell that it have an effect on plasticity of synapse and formation of neural network and lead to the descending of the learning and memory ability. Conclusion of the study is that the learning and memory ability disorders in rat after REM sleep deprivation may partially related to the expression abnormality of the number of nNOS positive cell and nNOS mRNA in rat cortex and hippocamp.
