Objectives:The defects of articular cartilage are an all too commonly encountered medical problem in bone and joint surgery,which may be caused by trauma or evolve during the course of diseases such as osteoarthritis,osteochondritis and osteonecrosis.Articular cartilage has a limited capality to repair spontaneously after injury. The large defects at articular cartilage are often repaired by fibrocartilage rather than normal hyaline cartilage. The resulting fibrocartilage are different from hyaline cartilage either biochemicallly or biomechanically and it ultimately deteriorates.It generally was thought that there was no effective clinical treatment that could restore a damaged cartilage surface and prevent the normal sequelae of degenerative joint disease.Recently, though there are many investigations to accelerate repairing of the defects of articular cartilage and improve the quality of repairing tissue,but no one satisfactory method and have a good long-term effect. The experiment`purpose is to study the effection of Chinese traditional medicine pilose antler growth factor on cell proliferationMethods: 1.To isolate the chondrocyte from a healthy adult rabbit bilateral hip and knee joint cartilage and then to culture;under sterilization2. To culture chondrocytes after 24 hours, and add pilose antler growth factor with 12mg/ml,8mg/ml,4mg/ml level respectively.3.To add 3H –TdR as a mark and measure the 3H -TdR scores in chondrocytes after 48,72 hours culture respectively, to observe the effection of DNA synthesis on chondrocytes of rabbit hip and knee joint in vitro . Results: 1.The group after added in pilose antler growth factor can improve cellular karyokinesis.2.After adding pilose antler growth factor with 48 hours, the 3H -TdR scores in chondrocytes is 2681±936 in superior group, 258±251 in median, 217±66 in small,compared with the control group 155±64, P<0.05.3.After adding pilose antler growth factor with 72 hours, the 3H -TdR scores in chondrocytes is 5592±2074 in superior group, 931±606 in median, 573±128 in small,compared with the control group 281±57, P<0.05.4.The 3H -TdR scores in chondrocytes after 72 hours culture is higher than that of 48 hours culture in the same level.Conclusions:The Chinese traditional medicine pilose antler growth factor can promote the proliferation of chondrocytes obviously, the effect vary in the different level.
