Objective To study the effect of herbal formula-gengniankang on different systems of aged female rats from immune-neuroendocrine system and bone, to explore the mechanisms of formula-gengniankang and to provide the important and scientific basis for the new treatment of menopausal syndrome. Methods1. Compared with adult female rats, aged female rats without oestrum were chosen by using RIA technology and sexual hormone was changed remarkably. Theses aged rats were divided into three groups(GNK group, HRT group and control group). Treated them respectively with GNK, nilestriol and distilled water 3 months. 2. Apoptosis of neuron in arcuate nucleus of hypothalamus was determined by TUNEL technology. To observe the effect of GNK on deterring aging process. 3. The level of serum E2,FSH and LH was determined by the method of RIA. Apoptosis of ovarian granulose cell were measured by TUNEL technology. To explore the effect of GNK on endocrine system. 4. Flow cytometry was used to evaluate the subsets and apoptosis of the spleen T cell. To study the effect of GNKon immune function. 5. The level of serum BGP and DPD was determined by the method of RIA and chemistry. BMD was measured by DEXA. RT-PCT technology was used to evaluate the expression of ER in bone. To observe the effect of GNKon bone from tissue level, metabolism level and mRNA level and to explore the mechanisms. Results1. Compared with adult female rats, apoptosis of neuron in arcuate nucleus of hypothalamus of aged female rats were increased(P<0.05). GNK can inhibit the process of apoptosis (P<0.05). 2. it occurred on aged rats that serum E2 decreased remarkably (P<0.05)and serum FSH,LH and apoptosis of ovarian granulose cell increased obviously(P<0.05). After treating with GNK, all the indexes were reversed except serum E2. The increase of E2 was not significant(P>0.05). 3. it occurred on aged rats that subsets of spleen T cell CD4+,CD4+/CD8+ decreased obviously(P<0.05) and apoptosis rate of T cell increased(P<0.05). After treating with GNK, all the indexes were reversed(P<0.05). 4. it occurred on aged rats that BMD and ER mRNA expression in bone decreased (P<0.05). These indexes were increased by GNK(P<0.05). Conclusion 1. GNK can inhibit further apoptosis of neuron in arcuate nucleus of hypothalamus, deterring aging process. 2. Apoptosis of ovarian granulose cell is decreased by GNK and it can deter further exhaustion of ovarian function. GNK regulates hormone of pituitary gonad axis. The fact that level of E2 in serum is not been increased should be pay more attention to. This suggests that GNK treating menopausal syndrome is not by increasing serum E2. 3. GNK can enhance the immune function of aged female rats and keep the balance of subsets of spleen T cell. 4. GNK is efficacious on aged female rats. The role is performed by enhancing the express of ER in bone, forming bone and increasing BMD.
