Objective To probe into the mechanism of derma and liver damage caused by trichloroethylene and do some immunological researches aim at dermatitis cases.Methods In Guinea Pig Maximization Test (GPMT),guinea pigs were immunized by intradermal(i.d.) vaccination with TCE(10%),second immunized by daub epidermal with TCE(20%) and challenged with TCE(10%).We use Flow Cytometry and immunohistochemistry detected subsets of lymphocyte of 8 dermatitis cases and we also use RT-PCR detected expression of gamma- interferon (IFN-γ) and alpha-tumor necrosis factor (TNF-α) mRNA of 5 dermatitis cases and 5 healthy controls.Results In TCE group, the ratio of immunization is 66%(33/50).Index of liver of dermatitis positive group increased to 3.89 (P<0.05). Alanine aminotransferase(ALT) increased to 128±52 IU/L (P<0.05),aspartate aminotransferase(AST) increased to 624±262 IU/L(P<0.05).Livers of dermatitis positive group showed severe deliquescence, nucleus of liver cells were not round,rough endoplasmic reticulum(RER) increased significantly. Dermatitis negative group have no signifacant change in these sides and the dosage of TCE used in research could not cause toxic liver damage.The results of flow cytometry indicated CD3+CD4+ was decreased (P<0.05),CD3+CD8+ was increased(P<0.05),CD+(16+56) and CD+19 was decreased significantly(P<0.001).There are no signifant difference between the results of two methods.The results of RT-PCR suggested the expression of IFN-γand TNF-α mRNA of dermatitis cases was up-regulated compared to those of healthy controls. Conclusion TCE can cause derma and liver damage with the dosage used in this research.The mechnism of derma damage may be corelation with the increase of CD3+ CD8+ and increase expression of cytokines such as IFN-γ and TNF-α.The mechnism of liver damage is under discussed.
