Aims: To investigate the effects of somatostatin analogue octreotide on the growth of gastric cancer in vivo.Methods:The animal models of xenotransplanted human gastric cancer cell line SGC-7901 in gymnomouse body were set up. Forty-eight nude mice were randomly divided into four groups: Octreotide , 5-FU, Octreotide plus 5-FU and control.All groups received medication respectively and then all were executed. The tumor mass volume .. weight, necrosis volume were observed and the inhibition rates for tumor growth were also caculated. The expressions of Ki67, VEGF Bcl-2 were assessed by Elivision plus immunohistochemical technique. Results: The average tumor volumes (cm3)in Octreotide group, 5-FU group Combined group and Control group was 2.17 0.78 2.19 0.79, 1.36 0.75, 3.23 0.74 respectively. The average tumor weight (g) was 6.88 2.06, 7.22 2.47, 4.47 2.28 , 10.30 2.80. The average tumor necrosis volume(cm3) was 0.55 0.38 0.52 0.53 , 1.14 0.83 , 0.32 0.27. The average tumor inhibition rate for Octreotide group, 5-FU group and Combined group was 33.20%, 29.92%, 56.60% respectively. The weakly positive express rates of Ki67 in Octreotide group, 5-FU group, Combined group and Control group were 50.0% 33.3% 25.0%, 0.0%; those of VEGF and Bcl-2 were 41.7%, 33.3% 8.3%, 16.7%;50.0% 41.7%, 16.7%, 8.3% respectively.The strongly positive rates of Ki67 VEGF , Bcl-2 in Octreotide group 5-FU group Combined group and Control groupwere 41.7%, 50.0% , 0.0% , 100.0%; 33.3% , 33.3% , 8.3%,83.3%; 16.7%, 25.0%, 0.0%, 91.7%;Conclutions: Octreotide could inhibit the growth of gastric cancer. Itsinhibition effect was the same as 5-Fu, Combined treatment withoctreotide and 5-Fu markedly enhanced the antitumor effect of 5-Fu.It could not only boost up the inhibition effects of 5-Fu, but alsomitigate the side effects of 5-Fu.
