| Objective: To study the effect of ShenFu on the expression of Bcl-2, Bax and c-myc in small intestinal cells after Ischemic Reperfusion injury in rats and explore its mechanismMethods: 36 SD rats weighing 225-275g were anaesthetized with ethyl urethane (Ig/kg), and rats were randomized into three groups each consisting of 12 rats: sham-operated control group(S), Ischemic Reperfusion+NS group (I/R +NS), Ischemic Reperfusion+ShenFu(I/R+SF), Ischemic Reperfusion models were made by occluded the anterior menseneric artery for 60 minutes followed by 120 minutes reperfusion,S and I/R+NS group were pretreated with Natrii Soldium 5ml/kg/h,and I/R+SF group were pretreated with ShenFu injection 5ml/kg/h.Immunohistochemical staining were detected the expression of Bcl-2, Bax and c-myc.TUNEL method measured apoptotic intestinal cells and calculated the apoptoic index.Results: 1. Histological evaluation S group: Normal. I/R+NS group: Epithelial cell necrosis,erosioned,Villi effaced,more neutrophils in lamina propria, glandular destruction. I/R+SF group: Epithelial cell degenerative changed but intact, mild inceased of lymphocytes and plasma cells in lamina propria. 2.The average OD value of Bax was significantly higher in I/R+NS group than that in S group(0.201 0.038 vs 0.116 0.026, P<0.01),but that was significantly lower in I/R+SF group than that in I/R+NS group(0.098 0.022 vs 0.201 0.038, P<0.01),and that was lower than in S group (0.098 0.022 vs 0.116 0.026, P<0.05). Compared with S group and I/R+SF group, the average OD value of Bcl-2 was higher and significantly higher(0.105 0.032 vs 0.084 0.015, PO.05; 0.145 0.030 vs 0.084 0.01, PO.01). and the average OD value of c-myc was significantly higher in I/R+NS group than that in S and I/R+SF group (0.088 0.011 vs 0.064 0.009; 0.088 0.011 vs 0.066 0.008, PO.01). The Bax> Bcl-2 and c-myc protein were located at cellular plasm, the positive cells had been stained yellow or brown.3.The apoptotic index in I/R+NS group was significantly higher than that in other two groups (25.325 1.323% vs 4.281 1.068%; 25.325 1.323% vs 5.946 1.623%, PO.01), and that in I/R+SF group was higher than that in S group(5.946 1.623% vs 4.281 1.068%, P<0.05).The positive cells stained by TUNEL assay had brown cellular nuclei with blue cellular plasm.Conclusion: ShenFu injection can enhance the expression of Bcl-2, decrease theexpression of Bax and c-myc, inhibit small intestinal cell apoptosis,protect small intestine injury after Ischemic Reperfusion. |
