[Objective]: In this study, we explore the change on expression of Fas and FasL of promoting apoptosis at different period of time after spinal cord injury (SCI) by making the model of transecting injury completely in SD rat and applying to the immunohistochemical method. We can reach to the aim that restrain the neuron apoptosis after SCI. This will open a new lookout for the theraphy after SCI. [Materials and Methods]: 30 healthy adult srague-daweley rats (female),whose mean weight 180-220g, were divided into 6 groups: normal control group, after spinal injury first day group, third day group, 7th day group, 14 th day group, 21th day group. After injecting Ketamine (l00mg/kg) and ansiolin (2.5mg/kg) by abdomen cavity, these animal were sacrificed via a way that infused 4% formaldehydum polymerisatum fluid into aorta with aortic cannulation. Then fixed the spinal cord segments (below the T10 spinal cord segments) taken out from the rats with 4% formaldehydum polymerisatum (4 ℃ And put them into 30% sucrose-PBS overnight ,lastly, these tissues were made into frozen section in 20μm thickness. Next,,anti-Fas and FasL serum were applied respectively with the immunohistochemical ABC method to detect the distribution and expression of Fas and FasL in spinal cord's gray matter of SD rats at different period time after SCI. The4grey degree of neuron in spinal anterior horn was measured by the analyzed system of image to determain the quantity of masculining reaction of Fas and FasL. [Result]: 1. In normal group and the kinds of the injured groups of the rat, Fas and FasL immunoreactivities were observed in anterior horn's motoneurons of spinal cord. 2. Immunoreactive expression of Fas and FasL varied at different period time after spinal cord completely transecting injury .(l)The number of Fas positive neuron of anterior horn's motoneurons in normal group had apparent chang compared with that of 1 day group, 3 days group, 7 days group, 21 days group (p<0.05), but it had no apparent chang compared with 14 days group (p>0.05). And, the number of Fas positive neuron of 1 day group, 3 days group and 7 days group increased apprarently than that of the normal group. However, the number of Fas positive neuron of 21 days group was lower than that of the normal group. The number of Fas positive neuron of 1 day group had no apparent chang compared with that of 3 days group (p>0.05), but that of 1 day group and 3 days group increased than that of 7 days group, 14 days group and 21 days group. The number of Fas positive neuron of 7 days group and 14 days group had apparent chang compared with that of 21 days group (p<0.05) and were higher than that of 21 days group.The compare of other groups had no apparent chang .(2) The average grey degree value of Fas positive neuron of the normal group had significant change compared with that of 1 day group, 3 days group and 7 days group (p<0.05), but that of the normal group had no significant change compared with that of 14 days group and 21 days group (p>0.05). The average grey degree value of Fas positive neuron of the normal group was higher than that of 1 day group, 3 days group and 7 days group. At same time, the average grey degree value of Fas positive neuron of 1 day group had no apparent change compared with that of 3 days group and 7 days group (p>0.05), but that of 1 day group had apparent change compared with that of 14 days group and 21 days group (p<0.05) and was less than that of 14 days group and 21 days group. The average grey degree value of Fas positive neuron of 3 days group had apparent change compared with that of 21 days group (p<0.05) and was less than that of 21 days group.
