Objectives: Through investigating the effect of acupuncture on weight, Lee's index, weight of fat tissue , the size of fat cells, the level of Leptin in celiac adipocytes, serum Leptin level and the contents of hypothalamic neuropeptide Y in obese rat fed by self-prescribed high calorie diet, research the mechanism of weight-loose function of acupuncture and provide a principle for the practice of acupuncture in clinic. Methods: Prepare the obese rats by self-prescribed high calorie diet. The serum Leptin and hypothalamic NPY were measured. Immunohistochemical analysis of Leptin in celiac adipocytes were performed. Results: Compared with obese rat without acupuncture, in acupuncture obese rat, the body weight and the wet weights of celiac fat were lower, Lee's index decreased, the adipocytes were smaller and the fat in the cells were lower, fibrous tissue increased inside of the cells, immunohistochemical stainings of Leptin were weaker, and the level of serum Leptin decreased, the hypothalamic neuroprptide Y level decreased. It suggested that the weight loose effect of acupuncture on obese rat was marked, and the machanism responsible for this was the affect of acupuncture on Leptin-hypothalamic neuropeptide Y chain. Conclusion: The effect of acupuncture on obese rat fed by self-prescribed high calorie diet was good, which may be obtained by its effect on Leptin- hypothalamic neuropeptide Y chain.
