Study On Expressive Spectrum Of PpGalNAcTs In Different Leukaemia Cells And The Effects Of Dissimilar Differentiation Revulsant

Objective: To observe the expression of polypeptide :N-acetylgalactosaminyltransferaseTs(ppGalNAcTs) in different leukaemia cells includingNB4 cell line,K562 cell line and SHI-1 cell line and to study the variation of ppGalNAcTsexpression after Chinese herbs saccarides and after l,25(OH)iD3 added to the culturemedium,to find out the relative function of ppGalNAcTs in the course of leukaemia'sgenerating and developing, as well as to make out the influence of Chinese herbssaccarides and 1,25(OH)2D3 on ppGalNAcTs.Methods: 1. Use RT-PCR to detect (1) the mRNA expression of ppGalNAcTs in NB4 cellline,K562 cell line and SHI-1 cell line;(2)the expression discrepancy of ppGalNAcTs inK562 cell line after Chinese herbs saccarides and 1,25(OH)2D3 added to the cultufemedium.2.Use MTT method to study the livability of K562 cells induced by Chinese herbssaccarides and by 1,25(OH)2D3.3.To study the apoptosis of K562 cells induced by Chinese herbs saccarides and by1,25-(OH)2D3 with the flow cytometer.Results: 1. The expression of ppGalNAc were different in three leukaemia cell lines.2. After Chinese herbs saccarides and after 1,25(OH)2D3 added to the culture medium,themRNA expressions of ppGalNAcT2,T4,T5,T7 in K562 cell line increased. The cell cycleHIwas arrested in Gl phase ;No cell death was detected.Conclusions: This study show that the expression of ppGalNAcTs in different leukaemia cell is different. This result suggests that different ppGalNAcTs have different particular substances.The expression of T2 exists in all cell lines which suggests it may be the key enzyme in O-glycan synthesis. The mRNA expression of ppGalNAcTs increases after the treatment of Chinese herbs saccarides and 1,25(OH)2D3.Which reveals that the identification and adhension function of cell strengthen and the vicious degree reduce.The results of Gl phase arrest and no cell death suggest the inhibition of cell growth because of not apoptosis but cell growth slower,which maybe indicate the begin of differentiation.
