Objectives: we select adult healthy New Zealand rabbits as our objects.Ligating/loosening the Left anterior descending branch (LAD)of the rabbit as the model of Ischemic Reperfusion( I/R ).we are to determine the protective effect of Puerarin(Pue) or (and) Ischemic Preconditioning (IPC) on myocardial Ischemic Reperfusion(I^R) injury,and to explore the molecular mechanism,by measuring the exponent of myocardial oxygen consumption,Creatine Kinase (CK), Lactate Dehydrogenase(LDH), Myocardial Water Content(MWC), concentration of GMP-140 in the plasma. et,al. Methods: 24 adult healthy New Zealand rabbits of either sex (2.6-3.2Kg,n=24) were randomly divided into four groups:the Control group(n=6), the Pue group(n=6), the IPC group(n=6) and the Pue+IPC group(n=6).the control group was subjected to only 30-min LAD occlusion followed by 120min reperfusion.The Pue group was given only Puerarin on l00mg/Kg and the IPC group only 5-min LAD occlusion followed by l0min reperfusion while the Pue+IPC group both,before they were subjected to 30-min LAD occlusion followed by 120min reperfusion. Those parameters of the exponent of myocardial oxygen consumption, CK,The Study on the Protective Effect of Puerarin Combining with IPC on Myocardial IRI________________AbatractLDH, Blood routine, MWC , concentration of GMP-140 in the plasma were measured after ischemic Preconditioning, the 30min, 60min, 120min of reperfusion. After that, Myocardial ultrastmcture changes of each group were observed. Results: The difference between the Pue+IPC group and the others ,the Pue group or the IPC group and the control group in the exponent of myocardial oxygen consumption, CK, LDH, MWC,concentra-tion of GMP-140 in the plasma were significant (P0.01 or P0.05). Conclusion: Ischemic reperfusion injury happened in all group. Puerarin or Ischemic preconditioning had their own myocardial protective effects. Combining Puerarin and Ischemic preconditioning as one protective measure ,we can get more powerful myocardial protective effects. Ischemic preconditioning has not only protective effects as Puerarin, but also its harmful side. Ischemic preconditioning doesn't give much effects on Heart Rate,while Puerarin can reduce the Heart Re.te and medial blood pressure significantly. Puerarin and Ischemic preconditioning both can depress the activation of platelet.
