Aurora kinases is a novel family of serine/threonine kinases that have been identified as key regulators of the mitotic cell division process including the regulation of centrosome function, bipolar spindle assembly and chromosome segregation progresses.Aurora-A is an important number of the serine/threonine kinases family and the kinase gene is located in the 20q13 chromosome region that is frequently amplified in many human cancers such as colorectal,breast,bladder and ovarian cancer.Aurora-A gene has been identified as a novel oncogene due to its overexpression detected in many human tumors as well as overexpression has been shown to chause tumorigenic transformation of human and rodent cells in vitro and in vivo. Lung cancer now ranks first among the malignancies that seviously threaten the human being's health and lives. Although all disciplines involved in lung cancer have advanced considerably,the incidence and mortality rate of lung cancer still continue to increase in recent years in China.Finding new therapeutic targets and prognosis markers are needed.A recent study has showed that aurora-A kinase is a potential molecular therapeutic target for pancreatic cancers.Another study reported that this gene may be a novel approach for non–Hodgkin's lymphoma. The expression of aurora-A kinase in lung cancer and associated research has not been reported since now.In the present study, the expression of aurora-A in lung cancer cell lines(A549,PG,NCI-H460) in mRNA,protein level was examined by semi-quantitative RT-PCR and Western-blot. The proportion of cells with 4N and >4N DNA content in three lung cancer cell lines were investigated by flow cytometry, the association between aurora-A expression and DNA content was analyzed,and this study provides a theoretical basis for finding a new prognosis marker and molecular therapeutic target for lung cancer.The result of semi-quantitative RT-PCR was analysis with TotalLab software,and the ratio of aurora-A/β-actin was 1.14(PG),1.16(A549), 0.84(NCI-H460),0.26(normal lung tissue). Aurora-A mRNA was overexpressed in all three lung cancer lines compared with normal lung tissue,and the level of aurora-A mRNA expression was different: A549>PG>NCI-H460.The result of western blot was also analysis with TotalLab software,and the ratio of aurora-A/β-actin was 21.13(A549), 6.43(NCI-H460),8.96(PG).Aurora-A was also overexpressed in three lung cancer cell lines,and the level of aurora-A protein expression were different:A549>PG>NCI-H460.The results show that aurora-A may possibly be a new lung cancer marker and molecular therapeutic target.The proportion of cells with 4N and >4N DNA content in A549,PG,NCI-H460 were 14.97%,19.88% and 10.6% respectively, and the difference of the proportion had statistical significance (p<0.01) .The proportion with >4N content were 3.59%,2.66%,2.30% respectively and A549 had significant difference with PG and NCI-H460 respectively (p<0.01); PG and NCI-H460 had no significant difference in the proportion (p>0.05). The proportion of PG with 4N DNA content was highest,which was related with the reason that it is a high-metastasis cell line.The proportion of A549 was higher than that of NCI-H460,which was concordant with the result of aurora-A expression.The rank of the proportion of cells with >4N DNA content were A549>PG>NCI-H460 that was concordant with the rank of aurora-A expression.So overexpression of aurora-A has corralation with the proportion of lung cancer cells with DNA content(>4N) .Because DNA content is a prognosis marker in lung cancer, aurora -A and expressed kinase may possibly be a new molecular marker to assess the prognosis of lung cancer. Aurora-A was overexpressed in three lung cancer cell lines,but the level of aurora-A mRNA expression were different. The 4N and >4N DNA content were different among three lung cancer cell lines.
