Objetive The gastric carcinoma is one of the most common malignant tumor in our country. The rate of incidence and death are highest in all kinds of the alimentary tract malignant tumors. The tumor cells invasion-metastasis and recurrence after surgery are the major reason for the poor prognosis of the patients with gastric carcinoma. Some study showed the selectins on the tumor cells and vascular endothelial cells closely related to invasion and metastasis of tumor cells. At present, many researches have reported that the expressions of P-and E-slectin in vein blood and carcinoma tissues of the patients with colorectal or breast cancer. However, There were no reports about the concentrations of the soluble P-and E-selectin in bone marrow of the patients with tumor, and few reports about their expressions in vein blood and carcinoma tissues of the patients with gastric carcinoma. By measuring the concentrations of the soluble P-and E-selectin in sera and marrow of the patients with gastric carcinoma and detecting the expressions of P-and E-slectin in gastric carcinoma tissues, we studied the correlation between the concentrations and expressions and the clinical biological characteristics of gastric carcinoma. The mechanism of gastric cancer cells invasion and metastasis is deduced from molecular biology level. It provided the molecular biological basis for the prediction of the invasion ,metastasis and progression of gastric cancer cells,the evaluation of the prognosis and treatment for the patients with gastric carcinoma. Methods A total of 72 patients with gastric carcinoma, proved by pathology, were recruited in the study. They were all come from He Bei Medical University Fourth Hospital Department 0f Surgery III. 72 cases of sera and marrow before surgery, surgically resected gastric carcinoma tissues and 40 cases normal gastric mucosa samples were collected, respectively. The patients or agents were told and signed before bone marrow penetrated. 32 of cases normal sera and 20 cases normal marrow samples derived from the heahty volunteers. The serum and marrow samples were measured for soluble P-and E-selectin levels by enzyme-linked immunosorbent assay (ELISA). The expressions of P-and E-selectin were detected on gastric carcinoma tissues and normal gastric mucosa specimens by immunohistochemical technique. At the same time, the correlations between the concentrations and expressions of P-and E-selectin and clinical biological characteristics, including sex, age, tumor location, size, differentiation, Borrmann type, depth of invasion, lymph node metastases and TNM stage, were analyzed. The data was organized and processed with SPSS10.0 statistical software. Results1 The concentrations of serum and marrow soluble P- selectin in the patients with gastric carcinoma were higher than those in the healthy people (p<0.01). But the concentrations of serum and marrow soluble E-selectin in the patients with gastric carcinoma were not significantly higher than those in the healthy people (p>0.05).2 The serum concentration of soluble P- selectin in the group with lymph node metastases was higher than that in the group without lymph node metastases (p<0.05). On the other hand, the concentrations of serum and marrow soluble P- selectin in the patients with gastric carcinoma were not correlated with sex, age, tumor location, size, invasive depth, differentiation, Borrmann type and TNM stage (p>0.05). The concentration of marrow soluble P- selectin in the patients with gastric cancer was not also related with lymph node metastases (p>0.05).3 The concentrations of serum and marrow soluble E- selectin in the group of tumor diameter ≧ 5cm, of low differentiated adnocarcenoma, of T3+T4, with lymph node metastase, of stage III-IV were higher comparing with those in the group of tumor diameter <5cm, high and moderate differentiated adnocarcinoma, of T1+T2, without lymph node metastases and of stage I-II, respectively.
