| Coleus forskohlii(Wild.) Briq.belongs to the Coleus genus of Labitae.The decoction of the plant is used in local folk medicine against cough,congestive cardiomyopathy,asthma,acute and chronic bronchitis, dizziness and fluster.Modern pharmacological researches have shown that Coleus forskohlii Briq.has good therapeutic efficacy against tumor metastasis,bronchial asthma,congestive heart failure and so on.We went on separating from active fraction and combined with the similar portion.Then got 10 fractions(Fra.A to Fra.J).Screened the anti-t umor activity fraction from the above fractions by MTT assay and Hel a cells used as test object.The results showed that the Fra.G,Fra.H an d Fra.J have significant anti-tumor activity.13 compounds from active fr actions were extracted and isolated by chromatography and identified 9 of them.Their structures have been determined as oleanolic acid(1),Lupeol(2),Uvalo(3),β-sitosterol(4),Colonic Acid(5),Demethylcryptoja ponol(6),Coleolic Acid(7),Coleon C(8),2-(2-Ethoxy-propyl)-3-hydroxy -5,7,8-trimethy- phenanthrene-1,4-dione(9).Lupeol(2),Uvalo(3),2-(2-Eth oxy-propyl)-3-hydroxy-5,7,8-trimethy-phenanthrene-1,4-dione(9) was found from HERBA LABITAE for the first time.2-(2-Ethoxy-propyl)-3-hydrox y-5,7,8-trimethy-phenanthrene-1,4-dione(9) was a new compound.The structures of compound(10),(11),(12) and(13) are being further identified.Screen the anti-tumor activity constituent from the above compounds by MTT assay and Hela cells and LS174-T cells used as test objects.oleanolic acid(1),lupeol(2),demethylcryptojaponol(6),Coleolic Acid(7),2-(2-Ethoxy-propyl)-3-hydroxy-5,7,8-trimethy-phenanthrene-1,4-dione(9) showed significant antitumor activity.To compare the micrology characteristics of tissues between wild and cultivated,Technologies based on tissue morphology are utilized to compare rhizomes micrology characteristic differences between wild and cultivated Coleus forskohlii Briq.The wild Coleus forskohlii Briq.is more obviously swollen than the cultivated at thrhizome because of more water-stockpiling parenchyma contained in the thrhizome of the wild.There are some micrology characteristic differences at rhizomes between wild and cultivated Coleus forskohlii Briq.,and this could present some forceful evidence for study, further exploiture and quality standard making. |
