Objective : To evaluate therapeutic effect of erythropoietin on hypoxia-ischemia encephalopathy (HIE) in neonatal rats and investigate the influence on them with different dose of EPO. Methods: 70 SD rats of 7-day old were divided randomly into sham-operated group(n=10), high dose group (5000IuAg n=20), low dose group (1000IuAg n=20) and experimental control group (normal saline n=20). Right after hypoxia-ischemia insult, EPO or normal saline were injected intraperitoneally and administered daily. Brain water contents were assayed in 48 hours and cerebral microscopical changes were observed on 7th day. Results Left brain water contents were (80.71 +0.69)%, (81.67+0.74)%, (82.61+1.07)%, the total number of neurocytes were 198.7+15.55, 156.6+9.55, 118.2+19.40, denatured necrotic neurons were 11.7+3. 50, 18.6+4.03, 24.40+3.44, respectively in high dose group, low dose group and experimental control group. Compared with experimental control group, left brain water contents , the total number of neurocytes and denatured necrotic neurons in EPO-treated group were different dramatically( all P<0.05 ),there are great difference between high dose group and low dose group too, indicating that EPO can attenuate HIE injury degree and brain edema ,andthe effect of high dose EPO is better than low dose therapeutically . Conclusion: EPO canexert therapeutic effect on HIE with dose-dependent manner.
