Objective:To investigate the regulation expresion of leukemia inhibitory factor(LIF) in human nomal and osteoarthritic (OA) articular tissue and related clinical changes. Methods : Human articular cartilage were obtained from operaion procedures and the good qulity RNA were isolated using Magnetic Beads with 32 frozen samples which were donored by osteoarthritise patients. Using the reverse transcription-polymerase chain reaction (RT-PCR), the mRNA express of LIF was messured by semi-qulitity, and the location of LIF protein was determimed by enzume-lmked immunosorbent assay(ELISA). We proved the related clinical symptoms,X-ray and histological study. Results (1) LIF mRNA was expressed to very low level in normal articular tissues and there was no significant difference (p=0.225) in different anatomical location.(2) In moderate degrading subchondral bone, LIF mRNA was expressed to the highest level. (3)LIF was expressed to the highest level in seriousely degrading articular cartilage tissue.This result similar with ELISA testing results.(4) LIF extents vary in different articular tissues section. Conclusions: (1) The different distribution of LIF in nomal and OA articular tissues may original from subchondral bone in the early period of OA. (2)Maybe it implys that LIF in coordination with other proinflammatory cytokine and plays the important roles in OA pathogenesis.
