Objective Cerebral stroke has already become the third most common cause of dead diseases, atherosclerotic thrombotic cerebral infarction(ATCI) is to above half of cerebral stroke.So prohibition from atherosclerosis remains a challenge. Recent studies have found that intercellular adhesion molecule-1(ICAM-1) plays a crucial role in lumphocyte migration and activation,and is considered important in the pathogenesis of atherosclerosis. K469E is a common polymorphism of the ICAM-lgene with potential functional significance.The aim of the present case-control study is to evaluate the association between this polymorphism and atherosclerotic thrombotic cerebral infarction.Methods we conducted a case-control study of 53 hospital cases with ATCI(male 36,female 17.mean age 63.58+7.71 years, age range 45-80 years) and randomly selected healthy control subjects without evident cerebral arterial disease(male 46,female 25.mean age 62.80+7.65, age range 45~80).Those who had skeptical informations of infection and embolism were excluded.The ICAM-1 K469E polymorphism was evaluated by polymerase chain reaction and restriction fragment length polymorphism(PCR-RFLP).TC,TG. and FBG levels were estimated by autoanalyzer.Results (1)In the ATCI group and control group the distribution of allele frequency was K: 0.37 and 0.24; E: 0.63 and 0.76, respectively.There was significant difference between them (x2=4.824, P=0.035) .In total specimen, allele frequencies of K and E were 0.29 and 0.71, respectively .The distribution of the ICAM-1 K469E polymorphism in above groups was EE genotype: 21% (11/53), 7%(5/71) ,respectively; EK genotype: 32% (17/53), 34% (24/71), respectively; KKgenotype: 47% (25/53), 59%(42/71) respectively. The EE genotype was significantly more common in patients than controls (P=0.031).The distribution of genotype frenquencies was in Hardy-Weinberg equilibrium.(2) Logistic regression analysis revealed that hypertension and diabetes mellitus were the independent risk factors of ATCI.Conclusions (1)The present study suggests that there is significant association beteewn the ICAM-1 K469E polymorphism and ATCI in Chinese Han ethnic group.(2) hypertension and diabetes mellitus are the independent risk factors of ATCI. |
