Study On The Mechanisms Of Anti-apoptosis And Anti-hypertrophy Of Kaixin Capsule In Left Ventricular Remodeling After Myocardial Infarction

Objective To explore the effect of kaixin capsule to apoptosis of cardiac musclecell, and expression of c-Fos,c-Jun, c-Myc and p21, p27 in left ventricular remodeling afterthe acute myocardial infarction. And to clarify the mechanisms of anti- apoptosis andhypertrophy of the decoction in left ventricular remodeling, and then to research thepositive accommodation of the capsule.Methods the experimental model of rat acute myocardial infarction wasstablished; the rats were feed by using Chinese traditional medicine decoction;the Captoprialgroup was given by Captoprial. the rats were killed 2 weeks after the treatment.Then, the rats' hearts were stripped, weighted, and fixed up with formaldehyde.To detect the expression intensity of c-Fos, c-Jun, c-Myc, p21 and p27 byperforming immunchistochemical technique, and the amounts of apoptosiscardiac muscle cell by using the TUNEL technique.Results1. Compared with artificial operation group, the number of apoptosis of cardiac muscle cell increased sharply in the blank control group and each treatment group. However, the number of apoptosis of cardiac muscle cell decreased in the treatment group when compared with the blank control group; and there weren't difference among the treatment group.2. Compared with artificial operation group, there were difference among the expression of c-Fos, c-Jun, c-Myc protein in the blank control group and-3-each treatment group. While, the expression of c-Fos c-Jun N c-Myc protein decreased in the treatment group when compared with the blank control group; and there weren't difference among the treatment group.3. Compared with artificial operation group, the level of c-Fos-. c-Jun% c-Myc protein of cardiac muscle cell decreased in the blank control group and each treatment group. However, the level of c-Fos c-Jun-. c-Myc protein of cardiac muscle cell increased in the treatment group when compared with the blank control group; and there weren't difference among the treatment group.Conclusion The kaixin capsule has obvious effect on anti-apoptosis andanti-hypertrophy. The mechanism of the decoction is likely to inducing theexpression intensity of c-Fos, c-Jun, c-Myc, increasing level of protein 21 and27, protecting the cardiac muscle cell.
