objective To study the significance and expression of p16,cyclin D1 and CDK4 in normal salivary gland and salivary gland carcinoma. Methods The expression of p16,.cyclin Dl and cdk4 in 10 normal salivary gland,30 mucoepidermoid carcinoma,and 30 adenoid cystic carcinoma were detected by immunohistochemical s-p assay. Results The expression rate of p1 6 in normal salivary gland and salivary gland carcinoma were 90% and 38% respectively. There was a significant difference between them (p<0.01). The expression rate of p16 had decreasing trend in salivary gland carcinoma. The expression rate of cyclinDl in normal salivary gland and salivary gland carcinoma were 20% and 80%respectively. There was a significant difference between them (p<0.01). The expression rate of cyclinDl had increasing trend in salivary gland carcinoma. The expression rate of cdk4 in normal salivary gland and salivary gland carcinoma were 30% and 88% respectively. There was a significant difference between them (p<0.01). The expression rate of cdk4 had increasing trend in salivary gland carcinoma. In salivary gland carcinoma, with the decreasing of the expression rate of p16, the expression rate of cyclinDl and cdk4 had increasing trend. An inverse correlation was found between the expression of p16 and cyclinDl, cdk4 (p<0.05). With the increasing of the expression rate of cyclinDl, the expression rate of cdk4 had increasing trend.An positive correlation was found between the expression of cyclinDl and cdk4 (p<0.05).P16 ,cyclinDl and cdk4 were not correlated with patients age and histological differentiation. Conclusions The decreasing expression of p1 6 may playan important role in the genesis of salivary gland carcinoma. The increasing expression of cyclinDl and cdk4 may play an important role in salivary gland carcinoma. Comprehensive analysis of p1 6, cyclinDl and cdk4 expression could help to understand the mechanism on salivary gland progression to malignancy.
