In the maintaining of the autoimmunological tolerance CD4+ CD25+ T cells play a major role. Removal of CD25+ cells, adoptive transferring CD4+ T cell to T cell-defecting mice, leads to various kinds of organ-specific autoimmune diseases, such as 1-type diabetes mellitus, glomerulonephritis, gastritis, polyarthritis, and so on. However, transfer of CD4+CD25+T cells and CD4+ T cells to mice, will prevent the occurrence of autoimmunediseases.Recently some investigations have revealed that, the functional character of CD4+CD25"IT cells is " anergic and suppressive". It is a focus of the correlation between CD4+ CD2s+ T cells and the episode of autoimmune diseases. In this text, we determinate CD4+ CD25+ T cells in anisocourse of systemic lupus erythematosus (SLE), to comprehend the expression of CD4+CD25+T cells in peripherial blood of patients with SLE, aiming to learn about the pathogenesis mechanism of SLE and find a new therapeutic method.Materials:53 patients with SLE, 14 normal controls with age- and sex-matched. According to SLE disease activity index (SLEDAI), we divided 53 SLE cases into active group (SLEDAI 10, 28 patients) and stationary group (SLEDAI < 10, 25 patients). Active group 1 (13 patients) never takes glucocorticoids and immunosuppressive drugs. Active group 2(15 patients) is taking glucocorticoids or immunosuppressive drugs. We have follow up 9 patients in active group 1 for 3 months. Flow-cytometry is used to determine CD4+CD25+T cells in peripherial blood.Methods:The levels of peripherical blood lymphocyte CD4+ CD25+ weremeasured by flow cytometry. Results:1 The expression of CD4+CD25+T cells in active group 1 is (17.58 8.10)%, lower than active group 2 (27.37 13.11)%, P = 0.019, stationary group ( 30.47 11.14 )%, P = 0.001 and normal control (38.23 9.37 )%, P =0.000. The expression of CD4+ CD25+ T cells in active group 2 is lower than normal control P = 0.009. The expression of CD4+ CD25+ T cells in stationary group is lower than normal control P = 0.035.2 The expressions of CD4+ CD25bright T cells in active group and stationary group are (2.42 2.13)% and (2.25 1.25)% respectively P = 0.727, which are significantly different from normal control (5.49 1.70)%, P=0.000, =0.000. The expressions of CD4+ CD25low T cells in active group and stationary group are (20.41 11.55)% and (28.23 10.92)% respectively (P = 0.011). There is the significant difference in the expression of CD4+ CD25low T cells in active group and normal control (32.74 卤 9.44)%, P = 0.001. There is no the significant difference in the expression of CD4+CD25lowT cells in stationary group and normal control, P = 0.220.3 The expressions of CD4+CD25brishtT cells and CD4+CD25lowT cells in active group 1 and active group 2: The expressions of CD4+CD25bnghtT cellsin active group 1 and active group 2 are (1.71 1.76)% and (3.03 2.29)%, u = 62.000, P = 0.108. The expressions of CD4+CD25low T cells in active group 1 and active group 2 are (15.87 7.10)% and (24.34 15.34)%, t = -2.046, P = 0.051.4 The expressions of CD4+ CD25bright T cells and CD4+CD25lowT cells in followed-up 9 patients who have never taken glucocorticoids or immunosuppressive drugs. The expressions of CD4+ CD2slow T cells are higher than that before therapy despite no significant differenc. But there is no change in the expressions of CD4+CD25bnghtT cells.5 Relationship between peripheral blood level of CD4+CD25brightT cells and CD4+ CD25low T cells and ANA titer, C3 and SLEDAI: It showed a correlation between the expression of CD4+CD25lowT cells and SLEDAI (p = -0.299, P = 0.030). There are no correlations between the expression of CD4+CD25brightT cells and ANA titer, C3 and SLEDAI.Conclusions:The expression of CD4+ CD25+ T cells in peripheral blood with SLE patients is decreased. The abnormal expression of CD4+ CD25bright T cells maybe correlated to pathogenesis of SLE.
