Objective To study the biological effect of L-arginine and Nitric Oxide process on the proliferation and differentiation of bone marrow stromal cells(BMCs). Method BMCs obtained by aspiration from rabbits, were incubated in DMEM medium and 100ml/L fatal bovine serum. Then the passaged cells were incubated with L-arginine with different concentration, in order to seek the most effective concentration on proliferation of the BMCs. Then the passage cells were divided into four groups: contrast group, L-arginine and dexamethasone group, and L-arginine or dexamethasone group. The ALP (alkaline phosphatase activity), MTT assay and stereological method were used to observe the effect on the proliferation and differentiation of BMCs. And NO generated by L-arginine was also investigated. Results L-argininecan improve the proliferation of BMCs, it also can reverse the proliferation inhibition in BMCs by dexamethasone(P<0.05). The cooperated group shows earlier and more in mount in cell differentiation than any other group(P<0.01). At same time, a correlation between these effects and NO was observed. Conclusions Dexamethasone combined with L-arginine can promotes the proliferation and differentiation, which can be related with the generation of NO.
