Objective To investigate the association between polymorphisms of both the 894G-T at exon 7 of endothelial nitric oxide synthase(eNOS) gene and the C-106T at promoter region of aldose reductase(AR) gene in chromosome 7q35 and nephropathy with type 2 diabetes of Han nationality in North China.Methods A case-control study for 202 Chinese subjects (including 139 type 2 diabetes mellitus with or without nephropathy and 63 normal control)was performed .The genotypes and alleles of polymorphisims of 894G -T at exon 7 of eNOS gene and C-106T at promotor region of AR gene were determined by polymerase chain reaction restriction fragment lenth polymorphism (PCR-RFLP) method and combined with 4% agarose gel electrophoresis. The results were validated by sequencing the PCR products.Results The frequency of the T allele and TG genotype at exon 7 of eNOS gene and the T allele and CT genotype at promoter region of AR gene were significantly higher in DN+ group than that in DN- group and control(CON) group (P<0. 05), but there was no significant difference between DM and CON group (P>0. 05).The type 2 diabetes with coexistence of the T allele/TG genotype of eNOS gene and T alleles/CT genotype of AR gene had higher incidence of diabetic nephropathy (P<0. 05) than those with only one of the two alleles/genotypes or without any of the two alleles/genotypes.Conclusion The T allele/TG genotype at exon 7 of eNOS gene and T alleles/CT genotype at promotor region of AR gene are related to nephropathy with type 2 diabetes of Han nationality in North China. The incidence of diabetic nephropathy is higher in patients who have both T allele / TG genotype of eNOS gene and T alleles/CT genotype of AR gene than those with only one of the two alleles/genotypes.
