Corneal disease is one of the most important disease which can cause blindness. Among corneal diseases, bacterial corneal ulcer and comeal thermal burn are seen frequently . According to many researches, Matrix metalloproteinases (MMPs) may have the important effect on bacterial corneal ulcer and corneal thermal burn . Researches have been trying to study the expression of MMPs and the mechanism and to look for the ideal inside and outside the tissue inhibitor of matrix metalloproteinases ( TIMPs) to decrease the damage . Now great progress have been made , but most of these research are focus on animal . The purpose of this experiment is to observe the expression of the MMP - 2 and the MMP - 9 in bacterial corneal ulcer and corneal thermal burn and to discover their functions in order to supply the necessary theory for the reasonable using of TIMP to prevent corneal damage.Materials and Methods1. Materials: we choose 10 healthy cornea and 10 cornea of bacterial corneal ulcer and 10 cornea of corneal thermal burn as experimental materials. The main reagents and equipments are rat monoclonal anti - people MMP - 2 and MMP - 9, peroxide enzymal goat anti - rabbit and anti - rat IgG, SP reagent box, computer analysis system.2. Methods; the corneal samples are saved in -80C. The corneas are tested in HE and Immunohistochemical dying. The expression of MMP - 2 and MMP - 9 is observed by microscop. The experimental results are analyzed by computer analysis system. Observing the change of MMP -2 and MMP -9 dur-ing different period of diseases.ResultsThe expression of MMP - 2 and MMp - 9 were found in the bacterial corne-al ulcer and corneal thermal burn and had statistics significance. MMP - 2 mainly located in corneal stroma. MMP - 9 located in corneal epithelium and basement membrane. The expression of MMP - 2 is different during the different period of the bacterial corneal ulcer. The case which is smaller than 30 days in the course of disease is relatively high, with the extension of the course of disease , MMP - 2 presents the downward trend. The change of MMP - 9 is not obvious ; In the course of the corneal thermal burn, MMP - 2 increase gradually at first and then drop; MMP - 9 express relatively high in initial stage and drop gradually on later stage.DiscussionBoth bacterial corneal ulcer and corneal thermal burn are destructive cornea disease that caused by cornea collagen degradation. MMPs plays a main role in this course. MMP - 2 and - 9 are the widest members among MMPs and play an important role in the bacterial corneal ulcer and corneal thermal burn.Our research shows in the bacterial corneal ulcer and corneal thermal burn, MMP - 2 and MMP - 9 positive expressing are relate to the different course of disease and the substrate, and there has a positive correlation between the quantity of expression and the degree of damage . With the extension of the course of disease, the expressions of two kinds of enzymes present the downward trend. MMP - 2 and MMP - 9 are of great importance in the course of the two diseases. It offered the preliminary theoretical foundation for possible medication of clinical practice. Because of the restriction of condition, we can not explain the expression of MMP -2 and MMP - 9 in the different period of the same disease and the same period of the two diseases, which are expected to offer more accurate theoretical foundation for clinical treatment.ConclusionMMP -2 and MMP -9 play an important role in the course of the bacterial corneal ulcer and corneal thermal burn, which are distributed with the substrate compatibly, and conformed with the degree and position of the pathological change.
