Objective: To observe the biological features of gastric cell SGC-7901 and the expression of HIF-1αduring hypoxia established in vitro and to study the mechanisms and effects of Genistein on gastric cancer cell SGC-7901 during hypoxia. Methods: The environment of hypoxia was established with GasPak method. MTT,FCM,transmission electron microscope,Cell-immuno- chemistry and the Boyden Chamber membrane invasion culture system were used to observe biological features of the gastric cells?the expression of HIF-1αand inhibitory effect of Genistein on SGC-7901 during hypoxia.Results: The PO2, PCO2 and pH remained stable during 48h in the case of GasPak method. There was no significant difference in the number of live cells between the normal state and hypoxia, and the cells were arrested in S phase. However,the motor ability, adhesiveness lamina basal and the expression of HIF-1αand TGF-β1 of SGC-7901 were strengthened during hypoxia. Genistein could inhibit significantly the growth of SGC-7901 both in normal and hypoxia state by arresting the cells in G2/M and G2/M-S phases and this inhibitory effect was elevated significantly during hypoxia. The electron microscope found the cell apoptosis and necrosis. The Fas expression were elevated by Genistein both in the normal and hypoxia states and the HIF-1αexpression were downregulated during hypoxia. Conclusion: GasPak method is an effective for cultivating gastric cancer cells in hypoxia because of its stability and easy replication. The expression of HIF-1αis elevated, and HIF-1αcan inhibit apoptosis of SGC-7901 cells during hypoxia. Gastric cancer cell SGC-7901 is tolerant to hypoxia. Hypoxia may lead to vicious conversion of the gastric cancer cell SGC-7901.Genistein can inhibit SGC-7901 cells growth by arresting the cell cycle, elevating Fas expression, inducing apoptosis and necrosis and other possible ways. Genistein can inhibit the expression of HIF-1αon SGC-7901 increased during hypoxia,perhaps it was important mechanisms of the inhibitory effect was elevated significantly during hypoxia.
