Appraisement Of Medicine And Treadmill Training On Ischaemia Stroke

Cerebral ischemia is a leading cause of death and serious, long-term disability in adults. Oxygen free radicaK energy metabolism apoptosis neurotransmitters etc, take part in the mechanisms of cerebral ischaemia. Medicine and execises are the main treatments of cerebral ischaemia.Using high performance liquid chromagraph microdialysis and flow cytometry etc,on the model of acute cerebral ischaemia and middle cerebral artery occlusion. the mechanisms of cerebral ischaemia were investigated and the appraisement of the effects of some medicine and exercises were taken.Using acute cerebral ischemia model in mice, the values of MDA LDH brain index (ratio of the brain weight with body weight ) and the activity- ATPase and Ca2 + - ATPase were observed, which reflected the procedure of dropsy, oxygen free radicaK energy metabolism, ions transporting, Ca2+ overloading, etc. Results showed that hawthorn leaves flavonoids (HLF) afforded protection against cerebral ischemia.Using HPLC and flurorescence detector, monoamino transmitters, such as NE, DA. 5-HT. and amino acid transmitters, such as Asp. Glu, Tau, GABA, were detected. By these methods, the changes of neurotranismitters in the brain of cerebral ischemia mice was detected and the effects of the medicine of Kedaling was appraised; By detecting the monoamino transmitters in the microdialysate of MCAO rats , the effect of Puerarin on anti-ischemia were appraised; And by detecting the amino acid transmitters in the culture medium of brain slices, the neuroprotective effects of Calmodulin(CaM) antagonist of trifluoperation(TFP) against hypoxic injury was appraised.Using Annexin V/PI assay, the apoptosis rate were detected in the cortex , striatum and hippocampus at different time. Results showed that the apoptosis rate first increased and then decreased .But the necrosis rate remained on some level.After MCAO rats'training on treadmill, their death rate declined, their behavirsand nerve function disfigurement were improved. But the infarc area of the brains were not decreased ,which proved that the mechanism of the acting of exercises lies in spinal cord, not in brain.
