Cell cycle is a dynamic process in which a cell experiences a stage from the past mitosis to the next mitosis, this process comprises 4 stages: G1, S, G2 and M. Cyclin-CDKS-CDKI have very important role in regulating this process. The imbalance of cell cycle or disorder of apoptosis is one of the important factors of many diseases. E2F is an important cell cycle regulator protein, which was discovered in 1986. It regulates the cell transition from G1 to S through Interaction with pRb and many other cyclin-dependent proteins. Rb, also called retinoblastoma protein, was the first discovered anti-oncogene. It presents as nonphosphorylationduring the period from GO to Gl, when it bind to E2F, it prohibits the transcription activation of E2F. P27, an multifunctional Cyclin-Dependent Kinase Inhibitor (CDKI) and one of main regulator of cell cycle, prohibits the cell proliferation through regulating the liminal value of cyclinE which is needed in the activation of CDK2.Skin Lichen planus is a subacute or chronic inflammatory skin disease, whose pathogeneses is unclear until now. Hyperkeratosis, granular layer hyperplasia, stratum spinosum irregularly Thickening, basal layer Liquefied degeneration, band like lymphocyte Infiltrate in dermis is a representative histological feature of lichen planus, which appears to may be related to an apoptosis reduction, cell hyperproliferation process. To try to find out the pathogeneses of lichen planus and comprehend the relation between the cycle regulatory proteins and the disease we detect the expression of E2F, Rb , P27 in skin lichen planus and discuss the significance of it.Material and method1, Source of the samples24 cases of LP were Biopsy samples of our department between March 1992 and November 2002. All cases were accord with clinical standard of LP and were diagnosed as LP in pathology. There were 12 male cases and 12 female cases, between 17 and 81 years old, the mean age was 48. 46 19.32. 11 cases were on lower extremity, 7 cases on trunk and 6 caseson upper extremity. 15 healthy controls were measured 2, Method:Rabbit anti-human E2F-1 polyclonal antibody KH95, rat anti-human Rb Monoclonal antibody Rbl, rat anti-human P27 Monoclonal antibody DCS-72.F6 (all were purchased from zhong shan biotechnology inc.) Immunohistochemistry was carried out using an Ultrusentivity鈩-P immunohistochemistry method, assays were performed accord to the manufacturer' s instruction. 3. Statistical analysis:All data was processed through SPSS11.5 kit. Difference between the LP and control group was analyzed with 2*3 tabularx 2 test. P value of less than 0.01 was considered significance.Result1. The expression of E2F in LPPositive cells for E2F were seen in control and LP conditions. However, a weak reaction for E2F in the basal and spinous cell layers were detected in control, 5 of 15 was stained positive, 33. 3 % was positive. In LP, expressions of E2F were prominent compared with those in the control group. And positive cells were seen in all layers, the most prominent was in spinous cell layers. 22 out of 24 were detected positive and 14 cases were stained strong positive. It was significantly higher in LP than it in control group (x2 =14.76 p=0. 001 ) .2. The expression of RbIn controll group Rb was strong positive, positive cells for Rb were seen in all layers in control conditions. However, a weak reaction for Rb was detected in LP conditions, and positive cells were sparse and scattered. 8 out of 24 was stained positive, 33. 3 % was positive. It was significantly lower in LP than it in control group( x2 =18.78 P<0. 001).3. The expression of P27 in LPThe expression of P27 in control epithelial was strong positive, positive cells for P27 were seen in all layers. Positive immunostaining was seen on the nuclear membrane and cell plasma. However, a weak reaction for P27 were detected in LP conditions,, positive cells distributed mainly in upper spinous cell layers, only nucleus immunostained. 12 out of 24 was stained positive,50...
