Effects Of QingningTang On The MRNA Expression Of MMP-2 And TIMP-2 In The Liver Tissuess Of Rats With Hepatic Fibrosis

Objective To investigate the effects of Qingning Tang on the mRNA expression of MMP-2 and TIMP-2 in the rats with experimental carbon tetrachloride-induced hepatic fibrosis ,and to explore the possible mechanism of Qingning Tang's anti-hepatic -fibrosis.Methods 168 SD rats were randomly divided into two groups :the normal control and the model groups.The hepatic fibrosis model was induced in the experimental rats by subscutaneous injection of 40% CCl4 3ml/kg/d body weight,twice a week for nine weeks, on the first time the dosage was double. After hepaticfibrosis model being built, the rats in the model group were divided into the hepatiofibrosis model, the high , the medium, the low dosage QingningTang and the Colchicine groups.Experiment disposal :The rats in the QingningTang groups were respectively given QingningTang at the dosage of 22g/kg/d ,llg/kg/d and 5.5g/kg/d ,the rats in the Colchicine group were given colchicines at the dosage of 0.5mg/kg/d,the rats in the normal control and the hepatic fibrosis model group were given the same volume saline,once a day for nine weeks.All the rats were killed at the end of the eighteenth week. The severity of inflammation and fibrosis in all liver tissuses were measured by semiquantitativescoring. At last,The mRNA expression of MMP-2 and TIMP-2 in liver tissues were detected by using in situ hybridization method.Results (1)Compared with the normal control group ,the severity of inflammation scoring and hepatic fibrosis scoring in the liver tissues of hepatic fibrosis model group was showed significant difference(P<0.01); Compared with the hepatic fibrosis model group, the severity of inflammation scoring and the liver fibrosis scoring in the liver tissues of all the QingningTang groups and the Colchicine group became obviously lower, and the severity of inflammation scoring in the liver tissues of the high dosage QingningTang group was markedly lowered, which showed significant difference(P<0.05,P<0.01). (2)Compared with the normal control group,the expression of MMP-2 mRNA in liver tissues of the other groups were obviously higher, which showed significant difference ; Compared with the hepatic fibrosis model group, the mRNA expression of MMP-2 in the liver tissues of Qingning Tang high , medium, low dosage groups show no significant influence (P>0.05 ), while the mRNA expression of TIMP-2 of liver tissues in the Colchicine groups showed significant difference (P<0.05). (3)Compared with the normal control group, the expression of TIMP-2 mRNA in liver tissues of the other groups were obviously higher, which showed significant difference(F<0.01) ;Compared with the hepatic fibrosis model group, the mRNA expression of TIMP-2 of the rats in QingningTang high , medium, low dosage groups werelower, which showed significant difference (P<0.05),while the mRNA expression of TIMP-2 of liver tissues in the Colchicine groups showed no significant difference(P>0.05).Conclusion CD QingningTang may reduce the severity of inflammation scoring and hepatic fibrosis scoring in the rats of experimental carbon tetrachloride-induced hepatic fibrosis . (2) The mRNA expression of MMP-2, TIMP-2 in the liver tissuess of the experimental hepatic fibrosis was enhenced,so MMP-2x TIMP-2 may be important factors in effecting hepatic fibrosis. (3)Qingning Tang may downregulate the mRNA expression of TIMP-2 in the liver tissuess of the experimental hepatic fibrosis,which may be the possible methanism of Qingning Tang's antihepatiofibrosis.
