Objective: To investigate the effect of celecoxib on inflammatory inhibition of septic rats. Methods: One hundred and forty-four SD rats were divided into three groups randomly. Group 1 received cecal ligation and punctures 1hr before normal saline was perfused into gastral cavity once a day; Group 2 received cecal ligation and punctures , and cephradine (0. 4g/kg)was perfused into gastral cavity Ihr later once a day;The third group received cecal ligation and punctures,and cephradine (0. 4g/kg)plus celecoxib(0. 0033g/kg) was perfused into gastral cavity 1h later once a day. The three groups sacrificed at 2hr, 4hr, 6hrs 1ds 3ds 7d after CLP, the plasma cytokine TNF- a which extracted from jugular vein was measured accordingly . The plasma cytokine IL-10 was measured at 8hrs 10hr. 12hr, 1d, 3ds 7d after CLP that was similar to TNF-a .The plasma ALB,GPT,LDH,BUN,Cr were determined before operation and 7 days later. The rats body weight were measured before operation and 7 days later likewise. Result: The cytokines TNF-a and IL-10 in the plasma of the 3 groups were increased by CLP. Celecoxib + cephradine groups decreased TNF-a and IL-10 level in plasma comparing to cephradine groups. Celecoxib caused the cytokines decreasing to some degree and attenuated the main organ inflammatory reaction , decreasing the excretion of urea nitrogen in septic rat. Conclusion: On the later phase of sepsis .Celecoxib could inhibit the release of cytokine , suppress inflammatory reaction, support the function of vital organ significantly. It is of great value in directing the clinic therapy of sepsis.
