Ischemia-reperfusion, which can injure tissue of the heart , bring difficulty for clinical treament of the heart disease . In order to seek cardioprotective drugs , we constructed the ischemia-reperfusion model of cultured myocardium of SD rat in vitro. Leakage of LDH of injured cardiac myocyte , content of oxygen free radical induced lipid peroxide metabolite-malondialdehyde(MDA) , activities of SOD(superoxide dismutase ), apoptosis ratio of myocardium injured by ischemia-reperfusion , the expression of related gene of apoptosis and effect of berberine on the above parameters were observed. Results: In ischemia, reperfusion group, the contents of LDH were 41.4IU/L and 51. 25IU/L separatively , compared with that of nomal group (22IU/L) have significant differences (P<0.01) ; contents of MDA in ischemia, reperfus ion group in supernatant were 15.2028 0.06 nmol/mL , 15.8997 0.20 nmol/mL ( P<0. 01, P<0. 01 respectively ), activities of SOD in the two group in supernatant were 13.4283 0.56 NU/ml , 11. 22 0. 29 NU/ml (P<0. 01, P<0. Olrespectively). Pretreatmentby berberine1.5 10-4mol/L, 1. 5 10-5mol/L and 1.5 10-6mol/L , respectively , the leakage of LDH in each group were all lower than that of non- pretreated group(P<0. 01), the contents of MDA(P<0. 01), the activities of SOD(P<0. 01). The apoptosis ratio of cardiomyocyte under ischemia, reperfusion situation were 26.4%, 41% much higher than central group 17. 4% (P<0. 05), In the two groups pretreated by berberine at 1. 5 10-5nmol/L, 1. 5 10-6nmol/L respectively, apoptosis ratio were all lower than non-pretreated group (P<0. 05), the expression of Bcl-2 protein in ischemia, reperfusion group decreased , which increased when pretreament by berberine (P<0.01) .Suggestions of results: berberine , a Chinese medicine , can stabilize mambrane of myocardium , restrict oxygen free radical injury , reduce ischemia-reperfusion injury . Berberine also improve expression of Bcl-2 protein in ischemia, reperfusion group, which prevent from occurance of apoptosis in cardiac myocytes during injury. The protective effects of berberine in part rely on its dosage within the range of concentration. It provides advantaged experimental basis for preventing and curing myocardium ischemia-reperfusion injury in usage of berberine .
