Abstract/Summary:
Objective Firstly ,to investigate the compatibility of PLA (polylactic acid) Glue in vitro which is a new kind of biomaterial. Secondly, to determine the ability of PLA Glue in the prevention of tendon adhesion. Lastly, to determine the ability of PLA Glue in the prevention of epidural scare adhesion after laminectomy.Methods (1)The tenocyte of chicken and the fibroblast of human were cultured with PLA Glue extract liquid of different concentrations and PLA Glue. The morphological changes and growths were observed respectively and the OD values of growths were evaluated by the MTT method. (2) The tendon of the soleus muscle of the left hind leg of the rabbit was partially separated and a transverse incision was made over half of its cross section. The incised site was then astoraosed and covered up with PLA Glue. In the control group, no of PLA Glue was used at the anastomosis site of the right hind leg. Gross and histologic observations were performed to evaluate the degrees of tendon adhesion at 2, 4 , 6 , 10 weeks .(3)The PLA Glue wereused in 18 rats using a total laminestomy model (L2) and not used in the rats of L5 (the control group). The gross anatomical appearance and microscopical evaluation were measured at 2, 4, 6, 8, 10, 12 weeks postoperatively.Results (1)Two kind of cells attached grew and proliferated well on the surface of material and in the PLA Glue extract liquid.. There were nosignificant difference in the OD value between these groups in which the cells were cultured with PLA Glue extract liquid of different concentrations and the negative control group. There were significant difference between the groups and the positive control group. (2) Gross and histologic studies showed that PLA Glue Significantly reduced the formation of the tendon adhesion.(3)Gross and histologic studies showed that PLA Glue significantly reduced the formation of the epidural scare. Thus the adhesion was prevented well.Conclusion The PLA Glue has very good compatibility and it is effective material to reduce tendon adhesion and duramater adhesion.
