Objetive To study the function of microsatellite replication error(RER) on chromosome 9 during the pathogenesis of acute leukemia and to investigate the significance of MSI and LOH in detecting the minima deletions of chromosome.Methods We have chosen 61 leukemia cases, including 33 cases of acute myeloblastic leukemia (AMD and 22 cases of acute lymphocytic leukemia(ALL) and 5 cases of chronic leukemia (CL), and 3 cases of myelodysplastic syndrome (MDS). MSI and LOH in 61 cases of leukemia were detected by multi-PCR amplification with their marrow cell and nomal oral mucosa cell at 5 microsatellites on chromosome 9, gel electrophoresis and argentine dye.Results 32 cases of acute leukemia demonstrated MSI at one or more loci.There were 15 cases found MSI and 10 cases found LOH in AML.In ALL the number is 5 cases and 4 cases respectively.The frequencies of LOH is 0(0\53) 5.7%(3/53) and 7.5%(4/53) at D9S162 , D9S1748 and D9S171 on 9p21 respectively.the frequencies of MSI in the three microsatellite is 15.1%(8/53), 7.6%(4/53) and 11.3%(6/53) respectively.LOH was found in 6 cases(11.3%) and 3 cases(5.6%) at D9S61 and D9S67 on 9q34 respectively.the frequencies of MSI at D9S61 and D9S67 is 5.6%(3/53) and 9.4%(5/53) respectively.6 cases detected LOH at D9S61 were all AML,the frequencies of LOH in AML(18.1%) weresignificantly higher than those of in ALL(0%).There were 6 cases(11.3%,RER+) found MSI at no less than two microsatellites.Furthermore,2 cases were found MSI in 5 CML cases.in which the two patient were in chronic and accelerate stage respectively.Only 1 cases was found MSI at D9S61 in 3 MDS cases.Conclusions The higher frequencies of RER+ in acute leukemia indicate that the pathogenesis and progress of acute leukemia is related to the extensive MSI;AML showed significantly higher LOH frequencies than ALL at D9S61. which suggest that there may be some gene alterations related to AML;The microsatellite tecnology is a new and valuable method to investigate leukemia.
