[Objective ] PTEN gene is a suppressor gene that dephospgorylates phosphatidylinositol 3,4,5-triphosphate,an intermediate in the PI3-K/Akt signaling pathway. It is known to be associated with tumour development in several human carcinomas. Studies on PTEN have improved since 1997. PTEN is a tumor suppressor which has sequence homology with dual-specficity phosphatase.lt is a multifunctional protein endowed with a phosphatase activity capable of dephosphorylating both tyrosine,serine/threonine phosphate residues on protein and phospholipids of the phosphatidylinositol pathway. PTEN appears to be mutated at considerable frequency in several types of human tumors,including those from brain,breast,endometrium,and prostate. PTEN play an important role in pathogenesis of tumor,tumor cell invasion and metastasis. In this review, we will investigate the expression of tumor suppressor gene PTEN protein to find the association between protein expression of PTEN and clinical pathological differentiation,stage andlymph nodes status of laryngeal carcinomas.[Methods] Using S-P, we studied 50 laryngeal squamous-cell carcinomas specimens and 40 vocal cord polyps with mouse monoclonal antibodys of PTEN .Disease stages were determined using the pTNM classification according to UICC criteria (1992). All specimens were approved by pathological examination after operation. All data were analyzed by Excel statistic software.[Results] We found that tumor suppressor gene PTEN was expressed lowly in laryngeal squamous-cell carcinomas (The rate was 62%).But the express rate was 100% in contrastThe differences were significant statistical!y(p<0.01).PTEN was expressed in highly, moderately, poorly differentiated squamous-cell carcinomas at frequencies of 82.4,57.1,20% respectively. The differences were significant statistically(p<0.05).Similarly, we found there were relativity between the clinical stages, lymph nodes status of laryngeal squamous-cell carcinomas and the expression of tumor suppressor gene PTEN protein(p<0.01).But it was no relativitybetween patients'gender,neoplasms'places,sizes and the expression of tumor suppressor gene PTEN protein.[Conclusion] So we thought that tumor suppressor gene PTEN expressed lowly in laryngeal squamous-cell carcinomas, and it is relative to pathological differentiation,stage and lymph nodes status of laryngeal carcinomas.Postgraduate: Liu Fu-xing (ENT)Directed by: Professor Zhao Shu you...
