IntroductionMutant p53 protein expression is one of biological items that reflect cellar reproduces activity . It has been gradually applied to the study of cardiology . No matter which kind of diseases, familiar HCM or sporadic HCM , it is not clear whether p53 protein involves in process of HCM etiology or not I. Therefore ,we examined the expression of p53 protein in peripheral blood lymphocyte in HCM group and control group by immunohistochemistry method, so that we can search the relationship between HCM and expression of p53 and provide a new theoretical idea for the study of HCMs etiology.Material and MethodsPatients were selected in China medical college affiliated No. 1 hospital and Liaoning province hospital from on april, 2001 to on de-cember, 2001 . there were totally 24 HCMs cases, who were verified by color echocardiogram and were divided further into LVOT obstruction subgroup and non - LVOT obstruction subgroup . there were 20cases in control group. They were tested that they did not suffer from hypertension, valvular heart disease, all kinds of cancer and leukemia through acquiring theirhistory, patients were taken 2 ml samples of peripheral vein blood, which were centriugated, and smeared, fixed, finally, these session were tested by immuunohistochemistry and staining method, when we judged results, we could see positive stained p53 protein that showed brown - yellow particles in nuclei, Occasionally, it can be seen in cytoplasm. Referring to the method that had used by Aoki and Singhams, we also thought about the percentage of positive cells in all same cells as well as staining" degree of positive cell at same time. They were respectively marked as ( - ),( +),( ++),( +++ ) . Judicial criteria was that ( - ) , ( + ) was reduced to negative expression cells, positive cells were marked as ( ++),( +++), while corresponding negative control cells werent stained. x2 test was adopted in the comparison between HCM group and control group. Exact probabilities in 2 x2 table was applied for the comparison between LVOT obstruction subgroup and non - LVOT obstruction subgroup, the relation between p53 proteins expression in the patients with HCM and the thickness of interventricular septum wall was judged by using rank correlation coeffieint.Results一,The comparison of p53 expression between HCM and control groupp53 positive expression rate of HCM group was 10% , that in the control group was 70. 8%. there was statistical difference between two group.二,the comparison of positive p53 protein expressionbetween LVOT obstruction subgroup and non - LVOT obstructionsubgroup .The positive expression rate of p53 protein in the patients with LVOT obstruction was 75.0% , that in the patients without LVOT was 62.5%. there wasnt statistical difference between two subgroups by u-sing exact probabilities in 2 x2 table.三,The relation between p53 protein's expression in the patients with HCM and their thickness of interventricular septum wallWe drawn the chart of p53 expression rate and corresponding thickness of IVS, then computed rs, ra = 0. 675. According to two sides test, we looked up the table of value r9, 0. 05(24) =0. 06 < 0.675,p <0. 05. It illustrated that the relation between p53 protein's expression in the patients with HCM and their thickness of interventricular septum wall was shown positive correlation in statistics.DiscusionBy now, the study of etiology had confirmed that 55% of the patients with HCM was related to autosomal genetic mutation. Sporadic patients with HCM , which were those patients that their parents hadn' t genetic abnormality, were 45% .At present, only did a few research report that a few patients were caused by b - MHC genetic mutation in sporadic HCM . major patients etiology were still unclear, it needed us to research further.Recently, Akira Matsumori found that sporadic HCMwas related with Hepatitis C virus(HCV) infection.
