Background And Objective:Being the core of the Hippo signaling molecules,MST1/2 and Yap1 have a promoting effect on the occurrence of tumor.This study aimed to investigate the expression and the clinical significance of Hippo pathway components MST1/2 and Yap1 in colorectal cancer(CRC).Materials And Methods:Eighty-eight cases of specimens were recruited from gastrointestinal endoscope room or surgical operation room in the second affiliated hospital of Nanchang university during June 2013 to December 2015.Among the 88,there were 30 colorectal cancer,30 human rectal adenocarcinoma,28 of benign colocrectal polyps.We used immunohistochemistry to detect the expression of MST1/2,Yap1 protein in the organization.According to the percentage of positive cells and expression intensity evaluation,we detected the expression of MST1/2,Yap1 protein and clinicopathologic features in colorectal adenocarcinoma.Evaluating the disease free survival period of colorectal cancer patients,we directed the relationship between Yap1 protein expression and prognosis of colorectal cancer.Results :The expression levels of MST1/2 protein in the colonic carcinoma group(median = 70)were significantly higher than those colon polyps group(median = 10),differences between the two groups was statistically significant(P<0.001);MST1/2 protein expression level in group rectal adenocarcinoma(median =60)obviously higher than that of colon polyps group(median = 10),the difference between the two groups was statistically significant(P = 0.004).The expression levels of Yap1 protein in the colonic carcinoma group(median = 50)obviously higher than that of colon polyps group(median = 0),differences between the two groups was statistically significant(P = 0.003).Yap1 protein expression level in group rectal adenocarcinoma(median = 55)obviously higher than that of colon polyps group(median = 0),the difference between the two groups was statistically significant(P<0.001).Furthermore,statistical analysis showed that there is nocorrelation(P>0.05)between the expression levels of MST1/2,Yap1 and the colorectal adenocarcinoma patients’ age,sex,smoking history,drinking history,location,tumor size,differentiation degree,T stage,N stage,tumor markers carcinoembryonic antigen(CEA)and carbohydrate antigen-199(Ca-199)expression level,blood test index level(WBC,RBC,HGB,PLT,ALB).The average DFS was 21.531 ±2.167 in the Yap1 positive group,and average DFS was 25.571 ±2.488 in the negative group;The mean value of DFS in the negative group is higher than the positive for 4 months,but no statistical difference was found between the two groups,(P=0.317);By comparing the survival curves of the two groups,the negative group is better than the positive group.Conclusion:Transcription activation factor-Yap1 may play an important role in colorectal adenocarcinoma development and cell malignant transformation.Yap1 may as an evaluation index short-term prognosis of recurrent colorectal cancer.Being as a tumor suppressor gene,the expression of MST1/2 in colorectal cancer tissue samples is lack of specificity,and subject to further study the imbalances in colorectal cancer.
